目的:研究绞股蓝正丁醇部位含药血清在体外对Aβ25-35蛋白诱导NG108-15细胞凋亡的抑制作用。方法:采用细胞免疫荧光术、流式细胞术检测绞股蓝正丁醇提取部位含药血清对Aβ25-35蛋白诱导NG108-15细胞凋亡的影响,以酶联免疫吸附法(ELISA)检测绞股蓝正丁醇提取部位含药血清对Aβ25-35蛋白诱导NG108-15细胞凋亡时胞内半胱天冬氨酸蛋白酶(Caspase-3)的水平的影响。结果:绞股蓝正丁醇部位含药血清在体外能明显抑制Aβ25-35蛋白诱导的NG108-15细胞凋亡,降低细胞内Caspase-3水平。结论:绞股蓝正丁醇部位含药血清在体外对Aβ25-35蛋白诱导的NG108-15细胞凋亡有一定的保护作用,其机制可能与降低NG108-15细胞内Caspase-3水平有关。
OBJECTIVE: To study the inhibitory effect of Aβ25-35 protein on the apoptosis of NG108-15 cells induced by drug-containing serum of n-butanol in Gynostemma pentaphyllum. Methods: Immunofluorescence and flow cytometry were used to detect the effect of serum containing n-butanol extract of Gynostemma pentaphyllum on the apoptosis of NG108-15 cells induced by Aβ25-35 protein. The effect of Gynostemma pentaphyllum Effect of medicated serum containing alcohol extract on the level of intracellular caspase-3 in Aβ25-35 induced apoptosis of NG108-15 cells. Results: The serum containing n-butanol in Gynostemma pentaphyllum could inhibit Aβ25-35 protein-induced apoptosis of NG108-15 cells in vitro and decrease the level of Caspase-3 in cells. CONCLUSION: The serum containing n-butanol in Gynostemma pentaphyllum can protect Aβ25-35-induced apoptosis of NG108-15 cells in vitro. The mechanism may be related to the decrease of Caspase-3 level in NG108-15 cells.
