采用聚酰胺-大孔树脂联用法纯化大叶藻黄酮,对其纯化工艺进行优化。样液以聚酰胺吸附,水洗除杂后置于大孔树脂柱上部,以70%(v/v)的乙醇洗脱。通过单因素和正交试验优化纯化的最佳工艺,确定最佳纯化工艺条件是样液与大孔树脂的比为4:1(v/m),样液与聚酰胺的比为16:1(v/m),洗脱体积为6 BV,洗脱流速为2 BV/h,在此条件下黄酮纯度由(14.97±0.51)%增加到(71.14±0.79)%。该方法纯化工艺简单,具有良好的应用前景。
Polyamide - macroporous resin was used to purify the flavonoids, and its purification process was optimized. The sample solution was adsorbed on polyamide, washed in water, removed from the column, and placed on the top of a macroporous resin column, and eluted with 70% (v / v) ethanol. The optimum conditions of purification were determined by single factor and orthogonal experiments. The optimum purification conditions were as follows: the ratio of sample solution to macroporous resin was 4: 1 (v / m); the ratio of sample solution to polyamide was 16: 1 (v / m). The elution volume was 6 BV and the elution flow rate was 2 BV / h. Under these conditions, the purity of flavonoids increased from (14.97 ± 0.51)% to (71.14 ± 0.79)%. The method has the advantages of simple purification process and good application prospect.
