目的探讨三氧化二砷(As2O3)抑制大肠癌SW620细胞增殖和转移的机制。方法将不同浓度的As2O3(0,1,2,4,8μmol/L)作用于人的大肠癌SW620细胞,用逆转录-聚合酶链反应(RT-PCR)检测各组大肠癌细胞NOTCH1mRNA表达,免疫细胞化学(SP法)检测各组CD133,KI67,血管内皮生长因子-C(VEGF-C)蛋白的表达。结果随着As2O3浓度的增加,NOTCH1mRNA、KI67、VEGF-C、CD133的表达逐渐减少,CD133与Notch1呈正相关,差异均有统计学意义(P<0.05)。结论 As2O3具有抑制大肠癌SW620细胞株细胞增殖和转移的作用,其机制可能与下调Notch1,以及对CD133、KI67、VEGF-C的抑制有关。
Objective To investigate the mechanism of arsenic trioxide (As2O3) inhibiting the proliferation and metastasis of human colorectal cancer cell line SW620. Methods Different concentrations of As2O3 (0,1,2,4,8μmol / L) were applied to human colorectal cancer SW620 cells. The expression of NOTCH1 mRNA in colorectal cancer cells was detected by RT-PCR, Immunocytochemistry (SP method) was used to detect the expression of CD133, KI67 and VEGF-C in each group. Results The expression of NOTCH1 mRNA, KI67, VEGF-C and CD133 gradually decreased with the increase of As2O3 concentration. There was a positive correlation between CD133 and Notch1 (P <0.05). Conclusion As2O3 can inhibit the proliferation and metastasis of human colorectal cancer cell line SW620, and its mechanism may be related to the down-regulation of Notch1 and the inhibition of CD133, KI67 and VEGF-C.
