目的:探讨止消通脉宁对自发性2型糖尿病KKAy小鼠肾纤维化及TIMP-1/MMP-2mRNA表达的影响。方法:将SPF级16周龄雄性KKAy小鼠50只按血糖值随机分为模型组、缬沙坦组、止消通脉宁高、中、低剂量组,另设10只C57bl/6J小鼠为正常组,连续灌喂给药,观察记录小鼠一般状况,4周后处死小鼠,取血和肾组织,检测血清BUN、Scr值,肾组织HE、PAS、Masson染色,用实时荧光定量PCR法检测肾组织中TIMP-1/MMP-2mRNA的相对表达量。结果:与模型组比较,止消通脉宁高剂量组尿蛋白有下降的趋势,肾功能得到改善,肾脏病理损伤减轻,肾组织TIMP-1/MMP-2mRNA水平均显著降低(P<0.05)。结论:中药复方止消通脉宁能够抑制自发性2型糖尿病KKAy小鼠肾组织TIMP-1/MMP-2mR-NA表达,减轻肾纤维化程度,这可能是其防治DN的作用机制之一。
Objective: To investigate the effects of Zhuxiaotongmaining on renal fibrosis and TIMP-1 / MMP-2 mRNA expression in spontaneously type 2 diabetic KKAy mice. Methods: Fifty male KKAy mice of SPF grade at 16 weeks old were randomly divided into model group, valsartan group and Zhuxiaotongmai high, medium and low dose groups according to the blood glucose level. Another 10 C57bl / 6J mice The mice were sacrificed after 4 weeks and the blood and kidney tissues were taken out. Serum BUN and Scr, renal tissue HE, PAS and Masson staining were detected, PCR method to detect the relative expression of TIMP-1 / MMP-2mRNA in renal tissue. Results: Compared with the model group, the urinary protein of Zhuxiaotongmaining high dose group showed a decreasing tendency, the renal function was improved, the renal pathological injury was relieved, and the level of TIMP-1 / MMP-2 mRNA in renal tissue was significantly decreased (P <0.05) . Conclusion: Zhuxintongmaining can inhibit the expression of TIMP-1 / MMP-2mR-NA in renal tissue and reduce the degree of renal fibrosis in spontaneously type 2 diabetic KKAy mice, which may be one of the mechanisms of its prevention and treatment of DN.
