目的 :检测抑癌基因PTEN蛋白表达在卵巢癌发生发展中的作用及相关病理机制。方法 :采用免疫组化法检测 95例卵巢癌组织、19例卵巢交界性肿瘤、10例正常卵巢、10例卵巢囊肿组织中PTEN蛋白表达 ,比较其表达与卵巢癌临床病理特征的关系。结果 :卵巢交界性肿瘤和卵巢癌组织中PTEN蛋白表达显著低于正常卵巢和卵巢囊肿 (P <0 .0 5 ) ;PTEN蛋白表达与卵巢癌组织分化程度呈正相关 (P <0 .0 1) ,与临床病理分期负相关 (P<0 .0 5 ) ;卵巢癌中PTEN蛋白表达降低或失活与卵巢癌淋巴结转移相关 (P <0 .0 1) ;上皮性卵巢癌中子宫内膜样癌PTEN蛋白表达最低 (P <0 .0 1)。结论 :在卵巢癌发生过程中PTEN蛋白表达降低起到重要作用 ,表达异常的PTEN蛋白与卵巢癌的组织分化程度、临床病理分期和转移能力相关 ,与上皮性卵巢癌中子宫内膜样癌PTEN蛋白表达关系更为密切。PTEN蛋白表达可作为判定卵巢癌发生及淋巴转移能力的一项有效客观指标。
Objective: To detect the role of tumor suppressor gene PTEN protein in the development of ovarian cancer and its related pathological mechanism. Methods: The expressions of PTEN protein in 95 cases of ovarian cancer tissues, 19 cases of ovarian borderline tumors, 10 cases of normal ovaries and 10 cases of ovarian cysts were detected by immunohistochemistry. The relationship between PTEN expression and clinicopathological features was analyzed. Results: The expression of PTEN protein in borderline ovarian tumors and ovarian cancer tissues was significantly lower than that in normal ovarian and ovarian cysts (P <0.05). PTEN protein expression was positively correlated with the degree of ovarian cancer differentiation (P0.01) , But negatively correlated with clinicopathologic stage (P <0.05). Decreased or inactivated PTEN protein expression correlated with lymphatic metastasis in ovarian cancer (P <0.01); in endometrial epithelial ovarian cancer The expression of PTEN protein was the lowest (P <0.01). Conclusions: PTEN protein expression plays an important role in the process of ovarian cancer. The abnormal expression of PTEN protein is correlated with the histological grade, clinical stage and metastasis of epithelial ovarian cancer, Protein expression more closely. PTEN protein expression can be used as a valid objective index to determine the occurrence and lymph node metastasis of ovarian cancer.
