目的 :探讨川芎嗪对活动期狼疮肾炎 (LN)外周血单个核细胞 (PBMC)的B7- 1(CD80 )和B7- 2 (CD86 )mRNA表达的影响 ,为临床使用川芎嗪治疗LN提供理论依据。方法 :取活动期LN患者PBMC培养 ,分别以川芎嗪、地塞米松和川芎嗪 +地塞米松进行处理 ,以PBS对照 ,采用半定量反转录PCR的方法 ,检测PBMC的B7- 1和B7- 2mR NA表达。结果 :与对照组相比 ,川芎嗪组和地塞米松组均能明显抑制B7- 2mRNA表达 (P <0 .0 5 ) ,而川芎嗪 +地塞米松组对B7- 2mRNA表达的抑制更加明显 (P <0 .0 1)。各组对B7- 1mRNA表达均无显著影响 (P >0 .0 5 )。结论 :川芎嗪能下调活动期LN外周血单个核细胞B7- 2mRNA表达 ,合并使用地塞米松能更好地抑制B7- 2mRNA表达。
Objective: To investigate the effect of ligustrazine on the expression of B7-1 (CD80) and B7-2 (CD86) mRNA in peripheral blood mononuclear cells (PBMC) of active lupus nephritis (LN) in order to provide a theoretical basis for the clinical use of ligustrazine in the treatment of LN . Methods: PBMC from patients with active LN were cultured and treated with ligustrazine, dexamethasone and ligustrazine respectively. Determine the expression of B7-1 and B7 in PBMCs by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) - 2mR NA expression. Results: Compared with the control group, both Ligustrazine group and dexamethasone group could significantly inhibit the expression of B7-2 mRNA (P <0.05), while the Ligustrazine + dexamethasone group inhibited B7-2 mRNA expression more significantly (P <0. 01). Each group had no significant effect on B7-1 mRNA expression (P> 0.05). CONCLUSION: Tetramethylpyrazine can down-regulate the expression of B7-2 mRNA in LN peripheral blood mononuclear cells during the active phase. Dexamethasone combined with dexamethasone can inhibit the expression of B7-2 mRNA.
