目的对分离纯化得到的3种大黄多糖结构进行研究。方法水提醇沉得到大黄粗多糖,分别经DEAE-52柱层析、Sephacryl S-200凝胶柱层析分离纯化后,得到3种多糖组分RP-1、RP-2、RP-3,并采用高效凝胶色谱GPC、薄层色谱TLC、气-质联用GC-MS、核磁共振NMR等对其分子量、单糖组成、糖链的连接方式及其分支情况和糖苷键构型等进行分析。结果 RP-1由葡萄糖残基构成,主要连接方式为1,4-连接,并在O-6位出现支链。RP-2由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖、半乳糖以及半乳糖醛酸组成,主要含有1,4-葡萄糖残基,同时还有1,2-鼠李糖,1,4,6-甘露糖、葡萄糖等残基。RP-3由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖以及葡萄糖醛酸组成,主要糖单元为1,5-连接的五碳糖。结论初步鉴定了3种大黄多糖的结构,为其药理作用的研究奠定了基础。
OBJECTIVE To study the structure of three kinds of rhubarb polysaccharides isolated and purified. Methods Rhubarb crude polysaccharides were obtained by water-alcohol precipitation. The polysaccharides RP-1, RP-2, RP-3 and RP-3 were obtained by DEAE-52 column chromatography and Sephacryl S-200 column chromatography respectively. The molecular weight, the composition of monosaccharides, the linkage of sugar chains and their branching and glycosidic bond configuration were determined by GPC, TLC, GC-MS and NMR. analysis. Results RP-1 consisted of glucose residues, the main linkage was 1,4-linkage and branched at O-6. RP-2 consists of rhamnose, arabinose, xylose, mannose, glucose, galactose and galacturonic acid and contains mainly 1,4-glucose residues, together with 1,2-rhamnose, 1 , 4,6-mannose, glucose and other residues. RP-3 consists of rhamnose, arabinose, xylose, mannose, glucose and glucuronic acid, with the main sugar unit being 1,5-linked pentose. Conclusion Preliminary identification of the structure of three kinds of rhubarb polysaccharides, which laid the foundation for the study of its pharmacological effects.
