目的探讨参芪扶正注射液对犬体外循环(CPB)缺血再灌注(I/R)肺细胞凋亡的影响及其可能机制。方法建立犬CPB模型,将实验犬随机分为CPB I/R组(对照组,n=6)和参芪扶正注射液治疗组(治疗组,n=6),在CPB前10 min(T1),主动脉开放后60 min(T2)、120 min(T3)在各相应时段采集同一肺叶肺组织1.0 cm3在同一固定液中固定,常规石蜡包埋切片,切片厚度为5μm,用SP免疫组织化学法检测Caspase-3、Bcl-2、Bax蛋白表达。结果在再灌注后60 min、120 min治疗组Caspase-3、Bax表达均较对照组减弱(P<0.05),Bcl-2表达则较对照组增强(P<0.05),Bcl-2/Bax较对照组增强(P<0.01)。结论在犬CPB过程中应用参芪扶正注射液,可以降低Caspase-3、Bax蛋白的表达,提高Bcl-2的表达,从而抑制肺泡细胞凋亡。
Objective To investigate the effect and its possible mechanism of Shenqi Fuzheng Injection on apoptosis of lung cells during cardiopulmonary bypass (CPB) ischemia-reperfusion (I / R) in dogs. Methods The canine CPB model was established. The dogs were randomly divided into CPB I / R group (n = 6) and Shenqi Fuzheng injection group (n = 6) (T2), 120 min (T3) after the aorta was opened. At the same time intervals, 1.0 cm3 of lung tissue of the same lobe was collected and fixed in the same fixative liquid. The paraffin sections were embedded in paraffin and the thickness was 5 μm. Results The expressions of Caspase-3 and Bax at 60 min and 120 min after reperfusion were weaker than those in control group (P <0.05), while the expression of Bcl-2 was significantly increased (P <0.05) and Bcl-2 / Bax Control group increased (P <0.01). Conclusions Shenqi Fuzheng injection can reduce the expression of Caspase-3 and Bax, increase the expression of Bcl-2 and inhibit the apoptosis of alveolar cells in canine CPB.
