目的 编制单细胞凝胶电泳 (SCGE)图像分析系统 (IMI 1.0 )。方法 利用Dephi 5作为工具 ,设计的分析指标有长度、强度、矩类以及头尾DNA含量比等 4类指标。结果 应用该系统检测了谷胱甘肽(GSH)在H2 O2 致DNA损伤中的作用 ,尾长、尾重心、Olive尾矩、尾惯量、尾分布矩、尾DNA含量 ( %)以及尾 /头比等指标与H2 O2 浓度存在明显的剂量 -效应关系。Olive尾矩与H2 O2 浓度的剂量 -效应关系线性良好 ,是SCGE检测DNA损伤的最适指标。 10 μmol/LGSH能抑制H2 O2 对DNA的断裂损伤。结论 IMI 1.0操作快捷方便 ,设计的指标能满足SCGE图像分析的要求。
Objective To develop single cell gel electrophoresis (SCGE) image analysis system (IMI 1.0). Methods Using Dephi 5 as a tool, the design analysis indicators include length, intensity, moment class, and ratio of head to tail DNA content. Results The system was used to detect the effect of glutathione (GSH) on DNA damage induced by H2O2. The tail length, tail center, Olive tail moment, tail inertia, tail distribution moment, tail DNA content (%) and tail / There was a significant dose-response relationship between H 2 O 2 and H 2 O 2 concentrations. The dose-response relationship between Olive tail moment and H2O2 concentration is linear and is the most suitable indicator of DNA damage by SCGE. 10 μmol / LGSH can inhibit the damage of H2 damage to DNA. Conclusion IMI 1.0 is quick and easy to operate, and the designed indexes can meet the requirements of SCGE image analysis.
