目的观察染尘细胞模型中人胚肺成纤维细胞(MRC-5)基质金属蛋白酶9(MMP9)及其组织抑制因子1(TIMP1)诱导表达的时间-效应关系。方法大鼠巨噬细胞(AM s)和MRC-5组成体外模型,采用逆转录多聚酶链反应(RT-PCR)技术检测体外染尘细胞模型中MRC-5分别在0,12,24,36,48,60 h等时段MMP9和TIMP1表达情况。结果 MMP9和TIMP1基因表达呈现明显时间-效应关系,S iO2染尘组相关系数r分别为0.639和0.976,2基因表达存在一定时差,早期以MMP9增高为主。结论基质金属蛋白酶生物合成的变化可能发生在转录水平。
Objective To observe the time-effect relationship of induced expression of matrix metalloproteinase 9 (MMP9) and tissue inhibitor of metalloproteinase-1 (TIMP1) in human embryo lung fibroblasts (MRC-5) in an infected cell model. Methods Rat macrophages (AM s) and MRC-5 were used as an in vitro model. MRC-5 was detected by reverse transcription polymerase chain reaction (RT-PCR) at 0,12,24,36, The expressions of MMP9 and TIMP1 in the same period of 48 and 60 h. Results The expression of MMP9 and TIMP1 showed a significant time-effect relationship. The correlation coefficient r of S iO2-exposed group was 0.639 and 0.976 respectively. There was a certain time difference between the expression of MMP9 and TIMP1. Conclusion The changes of matrix metalloproteinase biosynthesis may occur at the transcriptional level.
