目的:观察脓毒症大鼠急性肺损伤肺血管通透性、炎症因子、诱导型一氧化氮合酶(iNOS)的变化特点及其意义。方法:24只雄性SD大鼠随机分为脓毒症组和对照组,脓毒症组按5 mg/kg体重腹腔注射脂多糖(LPS),对照组给予等量生理盐水,于注射后2 h、6 h经腹主动脉采血、处死,比较动脉血气分析、ELISA法检测的肺组织iNOS、白介素-6(IL-6)含量、右下肺肺组织病理切片计算的病理学肺损伤积分。结果:与对照组比较,脓毒症组大鼠氧合指数显著下降、肺组织iNOS、IL-6含量、病理学肺损伤积分均显著增加,P<0.01。结论:脓毒症大鼠肺iNOS的过度表达增加了炎症因子的释放、增加肺血管通透性,加重肺损伤。
Objective: To observe the changes of pulmonary vascular permeability, inflammatory factors and inducible nitric oxide synthase (iNOS) in acute lung injury in sepsis rats and its significance. Methods: Twenty-four male SD rats were randomly divided into sepsis group and control group. Serum lipopolysaccharide (LPS) was injected intraperitoneally at a dose of 5 mg / kg body weight in sepsis group, and normal saline was given to the control group. After 2 h , 6 h after aorta blood sampling, sacrificed, compared arterial blood gas analysis, ELISA detection of lung tissue iNOS, interleukin-6 (IL-6) content, lower right lung lung histopathology calculated pathological lung injury score. Results: Compared with the control group, the oxygenation index of rats in sepsis group decreased significantly, and the scores of iNOS, IL-6 and pathological lung injury significantly increased, P <0.01. Conclusion: Overexpression of iNOS in lungs of septic rats increases the release of inflammatory factors, increases pulmonary vascular permeability and aggravates lung injury.
