目的 :观察卡氮芥 (BCNU)、威猛 (Vm - 2 6 )及尼莫地平 (NIM)联合应用对胶质瘤细胞的抑制作用 ,探讨联合用药剂量与胶质瘤细胞敏感性的关系 ,为临床提供有效的治疗依据。方法 :采用MTT法进行活细胞数测定及 [3H]-TdR掺入法观察药物对肿瘤细胞增殖率的影响 ,比较胶质瘤细胞对BCNU、Vm2 6及NIM单独使用、联合使用的敏感性。结果 :BCNU、Vm - 2 6和NIM联合使用对肿瘤细胞的抑制率可达 97.0 4% ,明显高于BCNU加NIM的抑制效果 (P <0 .0 1) ,也明显高于Vm - 2 6、NIM联合的抑瘤效果 (P <0 0 1)。同时BCNU、Vm - 2 6的浓度降低至单独使用的 1/ 10 - 1/ 10 0。结论 :BCNU、Vm - 2 6及NIM联合用药对肿瘤细胞的疗效明显优于单独用药 ,且具有抑制率高 ,用药浓度低的特点
OBJECTIVE: To observe the inhibitory effect of combination of BCNU, VIM - 2 6 and Nimodipine on glioma cells and to explore the relationship between the combination doses and the sensitivity of glioma cells. To provide an effective clinical basis for treatment. Methods: MTT assay was used to determine the number of viable cells and the incorporation of [3H] -TdR into tumor cells. The sensitivity of glioma cells to BCNU, Vm26 and NIM alone or in combination was compared. Results: The inhibitory rates of BCNU, Vm - 2 6 and NIM on tumor cells were 97.0 4%, which were significantly higher than those of BCNU plus NIM (P <0.01) and significantly higher than those of Vm - 2 6 , NIM combined anti-tumor effect (P <0 01). At the same time, the concentration of BCNU, Vm - 2 6 was reduced to 1/10 - 1/10 of that used alone. CONCLUSION: The combination of BCNU, Vm - 2 6 and NIM is superior to single drug on tumor cells, and has the characteristics of high inhibition and low drug concentration
