目的:探讨赤芍总苷对大鼠放射性肝纤维化的保护作用及机制。方法:雄性大鼠放射性肝纤维化动物模型,将大鼠随机分为正常对照组、模型组、赤芍总苷组(50 mg/kg、25mg/kg),除正常对照组外,其余各组均制备成放射性肝纤维化模型,造模后各组每天给予相应药物灌胃。于照射后第26周末剖杀大鼠,取血和肝脏标本,ELISA法检测大鼠中TGF-β1、TNF-α、IL-6、HA、PC-Ⅲ、LN含量,碱水法检测大鼠肝脏组织中Hyp含量;光镜下观察肝脏HE染色、MASSON染色病理改变;免疫组化法测肝脏组织TGF-β1、Smad3/4/7蛋白表达情况。结果:与正常对照组比较,模型组大鼠肝脏损伤和胶原纤维增生明显,血清中TGF-β1、TNF-α、IL-6、HA、PC-Ⅲ、LN含量明显增加(P<0.01),肝脏组织中Hyp含量增加(P<0.01),肝脏组织中TGF-β1、Smad3/4/7蛋白表达增加(P<0.01)。与模型组比较,赤芍总苷治疗组(50mg/kg)可显著降低肝纤维化大鼠血清中TGF-β1、TNF-α、IL-6、HA、PC-Ⅲ、LN的含量,同时可降低肝脏组织中Hyp含量及降低肝脏损伤程度及胶原纤维增生程度,还可减少大鼠脏肝组织中TGF-β1和Smad3/4蛋白表达,增加肝脏组织中Smad7蛋白表达。结论:赤芍总苷具有明显的抗肝纤维化作用,其机制可能与其阻断TGF-β1/Smad信号传导通路有关。
Objective: To investigate the protective effect and mechanism of total paeony glycoside on radiation-induced hepatic fibrosis in rats. Methods: The animal model of radiation-induced hepatic fibrosis in male rats was randomly divided into normal control group, model group and total paeoniflorin group (50 mg / kg, 25 mg / kg). Except the normal control group, Prepared into a model of radioactive liver fibrosis, each group after the model given the corresponding drug gavage. At the end of the 26th week after irradiation, the rats were sacrificed and the blood and liver samples were collected. The contents of TGF-β1, TNF-α, IL-6, HA, PC-Ⅲ and LN were measured by ELISA. Results: Compared with the normal control group, the liver injury and collagen fiber hyperplasia in the model group were significantly increased. The contents of TGF-β1, TNF-α, IL-6, HA, PC-Ⅲ and LN in the model group were significantly increased (P <0.01) The content of Hyp in liver increased (P <0.01), while the expression of TGF-β1 and Smad3 / 4/7 in liver increased (P <0.01). Compared with model group, TPG treatment group (50mg / kg) can significantly reduce the content of TGF-β1, TNF-α, IL-6, HA, PC-Ⅲ and LN in serum of rats with liver fibrosis, Reduce the content of Hyp in liver tissue and reduce the degree of hepatic injury and the degree of collagen fiber proliferation, but also reduce the expression of TGF-β1 and Smad3 / 4 in rat liver tissue and increase the expression of Smad7 in liver tissue. Conclusion: The total glycosides of Radix Paeoniae Rubra have obvious anti-hepatic fibrosis effect, and its mechanism may be related to the block of TGF-β1 / Smad signal transduction pathway.
