目的 探讨缺氧对肺癌细胞体外侵润力的影响及其分子基础。方法 将小细胞肺癌细胞H12 8分别置于常氧、低氧和无氧环境中培养 48h后 ,用羊膜内皮细胞模型测定各组细胞的侵润能力。同时测定并分析了上皮钙粘附素、β1 -整合素的表达量。结果 与常氧组相比 ,低氧组细胞侵润能力显著高于常氧组。低氧组E -钙粘附素表达显著降低及 β1 -整合素表达量显著增强 ,无氧组两者均显著降低。结论 适度缺氧可以下调肺癌细胞表达E -钙粘附素 ,增加 β1 -整合素表达 ,增强癌细胞侵润力。但是严重缺氧使癌细胞粘附分子表达及细胞侵润力均下降
Objective To investigate the effect of hypoxia on invasiveness of lung cancer cells in vitro and its molecular basis. Methods Small cell lung cancer cells H12 8 were cultured in normoxia, hypoxia and anaerobic environment respectively for 48 h. The ability of invasion of each group of cells was determined by amniotic endothelial cell model. At the same time, the expression of E - cadherin and β1 - integrin were measured and analyzed. Results Compared with normoxia group, the cell infiltration ability in hypoxia group was significantly higher than that in normoxia group. The expression of E - cadherin in hypoxia group was significantly decreased and the expression of β1 - integrin was significantly increased, both of which were significantly decreased in the anaerobic group. Conclusions Moderate hypoxia can down - regulate the expression of E - cadherin in lung cancer cells, increase the expression of β1 - integrin and enhance the invasiveness of cancer cells. However, severe hypoxia decreased the expression of cancer cell adhesion molecules and cell invasiveness
