目的 :从成年大鼠纹状体区和胎鼠皮层分离并鉴定神经干细胞。方法 :利用无血清培养和单细胞克隆技术在成年大鼠纹状体区和胎鼠皮层分离具有单细胞克隆能力的细胞群 ,选取单个克隆进行连续传代培养获得大量亚克隆 ,采用免疫荧光细胞化学技术检测克隆细胞Nestin抗原和分化后特异性成熟神经细胞抗原的表达。结果 :从纹状体区和皮层分离的细胞群具有连续克隆能力 ,胚胎早期细胞抗原 Nestin抗原阳性 ,分化后的细胞表达神经元、星形胶质细胞和少突胶质细胞的特异性抗原。结论 :我们分离的细胞具有自我更新能力和多分化潜能 ,属于胚胎早期细胞 ,有很强的增殖能力 ,是中枢神经系统的干细胞。
OBJECTIVE: To isolate and identify neural stem cells from adult rat striatum and fetal rat cortex. Methods: Cell populations with single cell cloning ability were isolated from adult rat striatum and fetal rat cortex by serum-free culture and single cell cloning technique. A single clone was selected for continuous subculture to obtain a large number of subclones. Immunofluorescence cytochemistry The technique was used to detect the expression of nestin antigen and differentiated specific mature neuronal antigen in cloned cells. RESULTS: The cell population isolated from the striatum and cortex had a continuous clonal capacity. Nestin antigen, an early embryonic cell antigen, was positive. The differentiated cells expressed neurons, astrocytes, and oligodendrocyte-specific antigens. Conclusion: The cells we isolated have self-renewal ability and multi-differentiation potential. They belong to early embryonic cells and have strong proliferative capacity and are central nervous system stem cells.
