水稻叶片中脉发育调控基因OsEL的克隆和功能研究

通过Affymetrix水稻表达芯片数据筛选出1个受低温和高温诱导高水平表达的基因OsEL(Erect Leaf,EL),该基因的蛋白序列含有SUI1保守结构域,可能为翻译起始因子蛋白。克隆OsEL基因并构建植物过表达载体,通过农杆菌介导转化水稻台北309,发现该基因的过量表达引起水稻株型发生变化,转基因植株的叶片变直立,叶片中脉变粗,中脉中的气腔和维管束数目增加,分蘖数增加。推测OsEL基因可能通过调控维管束细胞增殖影响叶脉的发育。 One Affymetrix expression-sensitive gene, OsEL (Erect Leaf, EL), was screened from Affymetrix cDNA microarray data. The protein sequence of the gene contains the SUI1 conserved domain, which may be a translation initiation factor protein. OsEL gene was cloned and overexpression vector was constructed. Agrobacterium tumefaciens-mediated transformation of rice in Taipei 309 revealed that the overexpression of the gene resulted in the change of rice plant type. The leaves of the transgenic plants became erect, the veins of the leaves became thick, and the air cavity in the midrib And the number of vascular bundles increased, the tiller number increased. It is speculated that the OsEL gene may affect the development of the veins by regulating the proliferation of vascular bundles.
