目的:观察双金痛立停胶囊对骨癌痛大鼠的作用及对肿瘤坏死因子α(TNF-α)含量的影响。方法:采用Walker 256乳腺癌细胞复制胫骨癌痛大鼠模型,选取模型复制成功的大鼠随机分为模型组、唑来膦酸(阳性药对照组)、双金痛立停胶囊19.6g,9.8g,4.9g生药/kg组,另同时设假手术组。测定各组大鼠给药前、后自由行走痛行为学评分、机械痛阈和热痛阈,末次给药后腹主动脉采血,运用酶联免疫法(ELISA)检测血清TNF-α的含量,观察骨组织的病理改变。结果:与模型组比较,双金痛立停胶囊19.6g生药/kg可明显降低骨癌痛大鼠的自由行走痛行为学评分,明显提高骨癌痛大鼠的机械痛阈值和热痛阈值,降低骨癌痛大鼠血清中TNF-α的含量,病理组织学检查表明,双金痛立停胶囊19.6g生药/kg能改善肿瘤引起的骨损伤。结论:双金痛立停胶囊对骨癌痛大鼠有镇痛作用,其机制可能与抑制肿瘤细胞分泌和释放TNF-α有关。
Objective: To observe the effect of Shuanghongdi Li Dong capsule on bone cancer pain rats and its effect on tumor necrosis factor α (TNF-α). METHODS: Walker 256 breast cancer cells were used to replicate the rat model of tibial cancer. The rats were randomly divided into model group, zoledronic acid (positive control group), Shuang Jin Tong Li Chang Capsule 19.6g, 9.8 g, 4.9g crude drug / kg group, while the other set sham operation group. The behavioral scores, mechanical pain thresholds and thermal pain thresholds were measured before and after administration of the rats in each group. Blood samples were collected from the abdominal aorta after the last administration, and the levels of serum TNF-α were detected by enzyme-linked immunosorbent assay (ELISA) Observe the pathological changes of bone tissue. Results: Compared with the model group, 19.6g crude drug / kg Shuangyinlongtiao Capsule significantly reduced the painless free-walking pain score of rats with bone cancer pain, significantly increased the mechanical pain threshold and thermal pain threshold of rats with bone cancer pain, Reduce the content of TNF-αin the serum of rats with bone cancer pain. The histopathological examination showed that 19.6g crude drug / kg of Shuangjinlongliyutang Capsule could improve tumor-induced bone injury. Conclusion: Shuangjin Tongliechan capsule has analgesic effect on bone cancer pain rats. The mechanism may be related to the inhibition of tumor cell secretion and release of TNF-α.
