目的:采用逆转录-聚合酶链反应(RT-PCR)法检测芍药苷预处理对皮质酮损伤的大鼠皮层神经元凋亡相关基因以及营养因子BDNF mRNA表达的影响,初步阐明芍药苷对皮质酮损伤神经元的保护机制。方法:体外培养胎鼠皮层神经元,于原代第7天加入芍药苷低、中、高浓度(0.5,2,10μmol.L-1)预处理30 min后,加入皮质酮(200μmol.L-1)损伤,采用RT-PCR检测芍药苷预处理对神经元凋亡相关基因Bcl-2,Bax,Caspase-3以及营养因子BDNF mRNA表达的影响。结果:与模型组比较,芍药苷预处理组(2,10μmol.L-1)可显著下调Bax,Caspase-3 mRNA的表达,并显著上调Bcl-2和BDNF mRNA的表达(P<0.05)。结论:芍药苷预处理对皮质酮损伤的大鼠皮层神经元的保护作用与芍药苷调控神经元凋亡相关基因Bcl-2,Bax,Caspase-3 mRNA的表达直接相关,同时可能与上调神经营养因子BDNF基因表达有关。
OBJECTIVE: To detect the effect of paeoniflorin preconditioning on cortical neuron apoptosis-related genes and BDNF mRNA expression in rat cortical neurons induced by corticosterone by reverse transcription-polymerase chain reaction (RT-PCR) Protection mechanism of ketone-damaged neurons. Methods: Cortical neurons were cultured in vitro. Preincubation of paeoniflorin with 0.5,2,10μmol·L-1 paeoniflorin for 30 min on the 7th day of primordial injection of corticosterone (200μmol.L- 1) injury. The effect of paeoniflorin preconditioning on the expression of Bcl-2, Bax, Caspase-3 and BDNF mRNA in neurons was detected by RT-PCR. Results: Compared with model group, Paeoniflorin pretreatment group (2, 10 μmol·L -1) significantly decreased the expression of Bax and Caspase-3 mRNA, and significantly up-regulated the expressions of Bcl-2 and BDNF mRNA (P <0.05). CONCLUSION: Paeoniflorin preconditioning has a direct protective effect on cortical neurons injured by corticosterone and the expression of Bcl-2, Bax and Caspase-3 mRNAs regulated by paeoniflorin in neurons of rats, and may be related to upregulation of neurotrophic Factor BDNF gene expression.
