目的:观察逍遥散对慢性应激损伤大鼠下丘脑室旁核促肾上腺皮质激素释放激素(CRH)表达的影响。方法:以逍遥散(5.265 g.kg-1)和糖皮质激素受体(GR)阻滞剂RU-38486(12.5 g.kg-1)为干预药物,应用慢性不可预知应激对大鼠进行为期3周的造模,在造模的同时给药。于第22天处死大鼠取材。采用原位杂交法测定各组大鼠下丘脑室旁核区CRHmRNA的表达。结果:逍遥散可显著下调慢性应激损伤大鼠下丘脑室旁核CRH阳性表达。结论:逍遥散可通过下调慢性应激损伤大鼠下丘脑室旁核区CRH mRNA表达而部分恢复慢性应激大鼠HPA轴负反馈功能。
Objective: To observe the effects of Xiaoyao powder on the expression of corticotropin-releasing hormone (CRH) in hypothalamic paraventricular nucleus of chronic stress rats. Methods: The rats were treated with Xiaoyao San (5.265 g.kg-1) and glucocorticoid receptor (GR) blocker RU-38486 (12.5 g.kg-1) A three-week modeling, while modeling at the same time. The rats were sacrificed on the 22nd day. The in situ hybridization was used to determine the expression of CRH mRNA in the hypothalamic paraventricular nucleus of rats in each group. Results: Xiaoyaosan significantly down-regulated the expression of CRH in paraventricular nucleus of hypothalamus in chronic stress rats. Conclusion: Xiaoyaosan can partially restore the negative feedback of HPA axis in chronic stress rats by down-regulating the expression of CRH mRNA in the paraventricular nucleus of hypothalamus in rats with chronic stress.
