从正常人血白细胞中提取基因组DNA,以此基因组DNA为模板,应用PCR方法扩增神经生长因子基因,把得到的基因片段重组到质粒pUC12上,经转化、筛选和鉴定得到含有中国人神经生长因子基因(β-NGF)的克隆。用双链末端终止法测定其全部354bp核苷酸顺序。测出的核苷酸顺序与文献报道的顺序有6处不同,据此推导出的氨基酸顺序有3处变化。推测中国人神经生长因子基因具有特殊性。
The genomic DNA was extracted from normal human leukocytes. The genomic DNA was used as a template to amplify the gene of nerve growth factor by PCR. The resulting gene fragment was recombined into the plasmid pUC12. After transformation, screening and identification, Cloning of Factor Gene (β-NGF). The entire 354 bp nucleotide sequence was determined by double-stranded terminator. There were 6 nucleotide sequences detected in the literature and 3 amino acid sequences deduced therefrom. Speculated that Chinese nerve growth factor gene has particularity.
