目的初步探讨双酚A对小鼠心脏组织的损伤作用。方法 50只健康昆明(KM)小鼠,按体重随机分为5组,每组10只,雌雄各半。剂量组分别给予2.4、24、240、480 mg/kg双酚A灌胃染毒,对照组给予玉米油灌胃,每天1次,共28 d。试验中计算心脏脏器系数,荧光分光光度法测定心肌组织中Ca2+-ATP酶的活性,通过HE染色观察心肌组织病理变化。结果与对照组比较,染毒组小鼠Ca2+-ATP酶活性下降(P<0.05),心脏脏器系数差异无统计学意义(P>0.05)。病理组织学检查可见心脏组织出现不同程度的改变,且随着剂量增加病理改变程度越显著。主要表现为心肌纤维排列疏松,间隙增宽,间质水肿,大量炎症细胞浸润,甚至心肌坏死。雌雄组之间无差异。结论双酚A对小鼠心脏组织可造成一定的损伤,其机制可能与双酚A导致小鼠心肌组织炎症与Ca2+-ATP酶活性下降有关。
Objective To investigate the effect of bisphenol A on cardiac tissue injury in mice. Methods Fifty healthy Kunming (KM) mice were randomly divided into 5 groups according to body weight, with 10 mice in each group. Dose groups were given 2.4,24,240,480 mg / kg bisphenol A intragastric administration, the control group was given corn oil gavage once a day for 28 days. Cardiac organ coefficient was calculated in the experiment. The activity of Ca2 + -ATPase in myocardial tissue was determined by fluorescence spectrophotometry. The pathological changes of myocardium were observed by HE staining. Results Compared with the control group, the activity of Ca2 + -ATPase in the exposed group was decreased (P <0.05), and there was no significant difference in the coefficient of cardiac organ between the two groups (P> 0.05). Histopathological examination revealed varying degrees of cardiac tissue changes, and with the dose increased the degree of pathological changes more significant. Mainly manifested as loosely arranged myocardial fibers, gaps widening, interstitial edema, a large number of inflammatory cell infiltration, and even myocardial necrosis. No difference between the male and female groups. Conclusion Bisphenol A can cause some damage to heart tissue in mice. The mechanism may be related to the inflammation of myocardial tissue induced by bisphenol A and the decrease of Ca2 + -ATPase activity in mice.
