研究妇科千金片对阿奇霉素(AZM)在慢性盆腔炎大鼠体内多次给药药物动力学的影响,评价联合用药的合理性。以雌性SD大鼠为实验对象,采用子宫内接种混合菌液加机械损伤法建立慢性盆腔炎动物模型。将模型大鼠随机分为对照组和联合用药组,对照组灌胃给予阿奇霉素10 mg·kg~(-1)·d~(-1),联合用药组给予等剂量阿奇霉素2 h后灌胃妇科千金片1.66 g·kg~(-1)·d~(-1),每日1次,连续7 d。于不同时间点采集血浆样本,HPLC-MS测定血浆中阿奇霉素浓度。采用ADAPT 5.1软件反向高斯模型计算药动学参数,SPSS软件对各组参数进行统计学分析。结果显示妇科千金片对阿奇霉素的药动学参数CL_t,CL_d,MIT,F均无显著影响,研究表明临床妇科千金片与阿奇霉素联合应用于慢性盆腔炎的治疗具有一定的合理性。
To study the effects of gynecology gold tablets on the pharmacokinetics of azithromycin (AZM) in rats with chronic pelvic inflammation and evaluate the rationality of combination therapy. Female Sprague-Dawley rats were used as experimental subjects. Animal models of chronic pelvic inflammatory disease were established by intrauterine inoculation of mixed bacterial liquid plus mechanical injury. The model rats were randomly divided into control group and combination group. The control group was orally given azithromycin 10 mg · kg -1 · d -1, and the combination group was treated with the same dose of azithromycin for 2 h. Gold tablets 1.66 g · kg -1 (-1) d -1, once a day for 7 days. Plasma samples were collected at different time points and the plasma concentrations of azithromycin were determined by HPLC-MS. Pharmacokinetic parameters were calculated using inverse Gaussian model of ADAPT 5.1 software, and SPSS software was used to analyze the parameters of each group. The results showed that gynecological Qianjin tablets azithromycin pharmacokinetic parameters CL_t, CL_d, MIT, F had no significant effect, studies have shown that clinical gynecology daughter gold tablets combined with azithromycin in the treatment of chronic pelvic inflammatory disease has some rationality.
