目的了解控制性超排卵(COH)对小鼠卵母细胞上表达水通道蛋白(AQP3)的影响。方法模拟人类超排卵周期,对小鼠进行超促排卵,按随机表顺序分为COH组和对照组,每组各10只ICR小鼠,用半定量实时荧光PCR方法检测小鼠MⅡ卵母细胞上AQP3mRNA的表达。结果卵母细胞膨胀实验显示,ICR小鼠卵母细胞上有AQP3的表达,用半定量实时荧光PCR方法检测时以β-肌动蛋白(β-actin)作为内参照,COH组和对照组ΔCT值分别是0·65±0·23和2·49±0·05,ΔΔCT值是1·84±0·18,两组比较P<0·001,具有统计学意义。结论控制性超排卵(COH)明显降低了ICR小鼠卵母细胞上AQP3mRNA的表达,这也可能是导致卵母细胞低冻融复苏率低的原因之一。
Objective To investigate the effect of controlled ovarian hypersexpression (COH) on the expression of aquaporin (AQP3) on mouse oocytes. Methods The human superovulation cycle was simulated and the ovarian hyperstimulation was performed in mice. The mice were randomly divided into COH group and control group, with 10 ICR mice in each group. Semi-quantitative real-time PCR was used to detect M Ⅱ oocyte On AQP3 mRNA expression. Results Oocyte swelling experiments showed that AQP3 was expressed on the oocytes of ICR mice. Β-actin was used as internal reference when detected by semi-quantitative real-time PCR. Values were 0.65 ± 0.23 and 2.49 ± 0.05 respectively, ΔΔCT value was 1.84 ± 0.18, P <0.001 in both groups, with statistical significance. Conclusions Controlled superovulation (COH) significantly reduces the expression of AQP3 mRNA in ICR mouse oocytes, which may be one of the reasons leading to the low freezing-thawing rate of oocytes.
