目的 探讨砷中毒的致癌机理及寻找灵敏、实用的 DNA损伤监测指标。方法 应用单细胞凝胶电泳法 (SCGE)对贵州省兴仁县燃煤型砷中毒重病区 175名砷接触者的血细胞 DNA损伤进行了检测 ,并分析了相关因素的影响。结果 砷能引起人体血细胞 DNA单链断裂 ,其损伤发生早于临床表现 ,并与尿砷、发砷呈正相关关系 :与皮肤损害进展一致。吸烟和氟的影响可加重砷对 DNA的损伤作用。结论 SCGE具有灵敏、快速、简便等优点 ,可作为一种早期、实用的方法应用于砷接触人群的 DNA损伤监测
Objective To investigate the carcinogenic mechanism of arsenic poisoning and to find a sensitive and practical indicator of DNA damage. Methods Single cell gel electrophoresis (SCGE) was used to detect DNA damage in blood cells of 175 arsenic-exposed patients in burned coal-borne arsenism in Xingren County, Guizhou Province. The effects of related factors were analyzed. Results Arsenic causes DNA single-strand breaks in human blood cells. The damage occurred earlier than clinical manifestations and was positively correlated with urinary arsenic and arsenic. It was consistent with the progress of skin lesions. The effects of smoking and fluoride can aggravate the damaging effects of arsenic on DNA. Conclusion SCGE is sensitive, rapid and simple, and it can be used as an early and practical method to detect DNA damage in people exposed to arsenic
