研究三七皂甙(PNS)对慢性缺氧大鼠肺动脉舒缩活动的影响。采用间断常压缺氧法制各大鼠慢性缺氧模型,腹腔注射(ip)PNS(100mg·kg-1·d-1)21天。3周后处死大鼠制备肺动脉血管环,观察乙酰胆碱(Ach)、钙离子载体A2318和硝普钠(SNP)对血管环的舒张作用。同时观察Ach和A23187对血管环一氧化氮(NO)生成的诱导作用及一氧化氮合成酶(NOS)抑制剂(L-NAME)对它们作用的影响。结果:PNS对正常大鼠离体肺动脉环产生剂量依赖的舒张作用,L-NAME和去内皮处理对PNS的这一作用无明显的影响。Ach、A23187和SNP对用PNS处理的缺氧大鼠肺动脉环诱导的舒张作用比正常大鼠和没有用PNS处理的缺氧大鼠强。Ach和A23187对用PNS处理的缺氧大鼠肺动脉环NO生成的诱导作用也较正常大鼠和没有用PNS处理的缺氧大鼠强。提示:①PNS对离体大鼠肺动脉有舒张作用,这一作用与NO无明显关系。②长期应用PNS能增强缺氧大鼠肺动脉内皮细胞依赖的舒张反应和NO的生成。
To study the effects of panax notoginseng saponins (PNS) on the pulmonary vasomotion of chronic hypoxic rats. Chronic hypoxia model was established by interrupted atmospheric hypoxia method. PNS (100 mg·kg-1·d-1) was intraperitoneally injected (ip) for 21 days. Pulmonary arterial vascular rings were prepared after sacrificed 3 weeks. The relaxation effects of acetylcholine (Ach), calcium ionophore A2318 and sodium nitroprusside (SNP) on the vascular rings were observed. The effects of Ach and A23187 on the induction of nitric oxide (NO) production in vascular rings and the effects of nitric oxide synthase (NOS) inhibitors (L-NAME) on them were also observed. RESULTS: PNS exerted a dose-dependent relaxing effect on isolated pulmonary rings of normal rats. L-NAME and deendothelialization had no significant effect on PNS. Ach, A23187, and SNPs induced relaxation of the pulmonary arterial rings in hypoxic rats treated with PNS more strongly than normal rats and hypoxic rats that did not receive PNS. Ach and A23187 also had stronger induction of NO production in Pulmonary artery rings of rats treated with PNS than normal rats and hypoxic rats that were not treated with PNS. Tip: 1PNS has a relaxing effect on pulmonary arteries in isolated rats. This effect is not significantly related to NO. 2 Long-term application of PNS can enhance pulmonary artery endothelial cell-dependent relaxation response and NO production in hypoxic rats.
