为探讨衰老过程中转录活性水平下降的机理,用同位素掺入法和脱氧核糖核酸酶Ⅰ(DeoxyribonucleasI,DNaseⅠ)作为检测基因活性的探针,对人胚肺成纤维细胞2BS在衰老过程中染色质转录活性的变化进行研究。衰老细胞核染色质体外转录活性较年轻细胞下降58%,并对DNaseI的敏感性也较年轻组有明显降低。提示:衰老2BS核染色质结构的改变可能是转录活性改变的重要因素。
In order to explore the mechanism of the decline of transcriptional activity in the process of aging, isotope incorporation and Deoxyribonucleic acid Ⅰ (DNase Ⅰ) were used as probes to detect gene activity. Chromatin Changes in transcriptional activity were investigated. Senescent nuclear chromatin in vitro transcription activity decreased 58% compared with young cells, and DNaseI sensitivity than the younger group was significantly lower. Tip: Changes in the chromatin structure of aging 2BS nucleus may be an important factor in the transcriptional change.
