目的:研究不同剂量的甲基苯丙胺(MA)对小鼠条件性位置偏爱(CPP)的影响以及左旋四氢巴马汀(l-THP)自身是否产生CPP及对MA诱导的CPP的影响,并初步探讨其神经生物学机制。方法:本实验采用CPP倾向性实验程序,以C57BL/6小鼠为对象腹腔注射(ip)MA,小鼠随机分为8组,即NS组,0.25mg·kg-1MA组,0.5mg·kg-1MA组,1.0mg·kg-1MA组,2.0mg·kg-1MA组,5.0mg·kg-1MA组,10.0mg·kg-1l-THP组,1.0mg·kg-1MA+10.0mg·kg-1l-THP组,各组小鼠先前测1d,训练8d,d10测试各组小鼠对不同处理的CPP的表达。结果:(1)0.5mg·kg-1,1.0mg·kg-1,2.0mg·kg-1的MA能诱导出CPP效应(P<0.05),而且1.0mgkg-1的MA诱导的CPP效应最明显(P<0.01);(2)10.0mg·kg-1的l-THP自身不能诱导CPP效应;(3)后测前30min给予10.0mg·kg-1的l-THP能明显抑制1.0mg·kg-1MA诱导的CPP表达(P<0.01)。结论:(1)不同剂量的MA(0.25mg·kg-1-5.0mg·kg-1)所诱导的CPP效应并不成单一的量效关系;(2)l-THP自身没有奖赏效应;(3)l-THP对MA的奖赏效应有一定的抑制作用。
AIM: To investigate the effects of different doses of methamphetamine (MA) on conditioned place preference (CPP) in mice and the effects of l-tetrahydropalmatine (l-THP) on the production of CPP and MA-induced CPP Preliminary study of its neurobiological mechanism. Methods: In this experiment, CPP propensity experimental procedure was used to intraperitoneally inoculate C57BL / 6 mice with MA. The mice were randomly divided into 8 groups: NS group, 0.25mg · kg-1MA group and 0.5mg · kg -1MA group, 1.0mg · kg-1MA group, 2.0mg · kg-1MA group, 5.0mg · kg-1MA group, 10.0mg · kg-1l-THP group and 1.0mg · kg-1MA + 10.0mg · kg- 1l-THP group. The mice in each group were measured for 1d and trained for 8d. The expression of CPP in different groups was tested by d10. Results: (1) CPP was induced by 0.5mg · kg-1, 1.0mg · kg-1 and 2.0mg · kg-1 MA (P <0.05) (2) 10.0 mg · kg-1 of l-THP could not induce CPP by itself; (3) 10.0 mg · kg-1 of l-THP 30 min before the test significantly inhibited 1.0 mg · kg- kg-1MA induced CPP expression (P <0.01). Conclusions: (1) CPP effect induced by different doses of MA (0.25mg · kg-1-5.0mg · kg-1) does not have a single dose-effect relationship; (2) l-THP has no reward effect by itself; ) l-THP has a certain inhibitory effect on the reward effect of MA.
