目的研究玫瑰花和鲍鱼菇具有抗氧化活性的提取物对实验性糖尿病小鼠的降血糖作用机制。方法RT-PCR方法检测玫瑰花和鲍鱼菇提取物对糖尿病小鼠胰腺胰岛素、超氧化物歧化酶(SOD)、胰岛素激活因子(PDX-1)基因表达水平的影响。结果两种提取物均能够明显提高各基因的表达水平,玫瑰花和鲍鱼菇提取物组的胰腺胰岛素基因表达分别比糖尿病模型组提高22.2%和17.6%,SOD基因表达提高20.1%和9.3%,PDX-1分别提高19.1%和27.4%。结论玫瑰花和鲍鱼菇提取物均能够明显提高各基因的表达水平,通过提高胰腺的抗氧化能力发挥降血糖作用,但降血糖作用机制不同。
Objective To study the mechanism of hypoglycemic effect of extracts with antioxidant activity of rose and abalone mushroom on experimental diabetic mice. Methods RT-PCR method was used to detect the effect of rose and abalone mushroom extract on insulin, superoxide dismutase (SOD) and insulin activating factor (PDX-1) gene expression in diabetic mice. Results Both extracts could significantly increase the expression level of each gene. The pancreatic insulin expression in the rose and abalone mushroom extract groups was 22.2% and 17.6% higher than that in the diabetic model group, and the SOD gene expression was increased by 20.1% and 9.3%. PDX-1 increased by 19.1% and 27.4%, respectively. Conclusion Both rose and abalone mushroom extracts can significantly increase the expression level of each gene and play a hypoglycemic effect by increasing the antioxidant capacity of the pancreas, but the mechanism of hypoglycemic action is different.
