目的探讨骨髓间充质干细胞(MSCs)对造血干细胞(HSCs)“干性”的保持作用及其机制。方法免疫磁珠分离纯化脐血HSCs-CD34+细胞,并分组培养:A组,CD34+细胞单独培养;B组,加细胞因子;C组,加细胞因子和MSCs(直接接触);D组,加细胞因子和MSCs(不接触);E组,加MSCs(直接接触);F组,加MSCs(不接触)。0、7、14d分别计扩增倍数、单位细胞数形成集落能力、CD34+比率,计数7d时各组CD34+、CD34+CD38-比率。结果随着时间延长扩增倍数增加(尤其B组),形成集落的能力和CD34+细胞比率渐下降,但E、F组下降不明显。结论以MSCs为滋养层,对HSCs扩增作用不明显,但可更好地保持HSCs的“干性”;这种保持“干性”的作用依赖于细胞与细胞间的相互黏附作用。
Objective To investigate the effect and mechanism of mesenchymal stem cells (MSCs) on maintenance of hematopoietic stem cells (HSCs). Methods The cord blood HSCs-CD34 + cells were isolated and purified by magnetic beads. Group A: CD34 + cells were cultured alone; group B: cytokines were added; group C: cytokines and MSCs were added; Factor and MSCs (no contact); Group E, plus MSCs (direct contact); Group F, plus MSCs (no contact). 0, 7, 14d, respectively. The number of cells in each group formed colony-forming ability, CD34 + ratio, CD34 +, CD34 + CD38- ratio in each group at 7d. Results With the extension of time, the amplification fold increased (especially in group B), the ability to form colonies and the percentage of CD34 + cells gradually decreased, but the decrease in groups E and F was not obvious. CONCLUSION MSCs as a trophoblast have no obvious effect on the HSCs proliferation, but they can better maintain the “dryness” of HSCs. The function of maintaining “dryness” depends on the mutual adhesion of cells and cells effect.
