目的优化及评价异体抗原联合醋酸诱导溃疡性结肠炎大鼠模型的建立方法。方法以不同的抗原乳化液蛋白含量、不同醋酸浓度及醋酸在大鼠体内滞留时间作为变量,根据大鼠的疾病活动指数(DAI)评分和结肠黏膜损伤指数(CMDI)评分,筛选出最优的蛋白浓度、醋酸浓度及醋酸在大鼠体内滞留时间组合。结果固定醋酸体内滞留时间为13 s,以醋酸浓度和抗原乳化液蛋白用量为变量进行三水平的正交实验。抗原乳化液蛋白用量为0.1m L浓度80 mg/m L,5%醋酸溶液1 m L的造模组与正常组相比,DAI评分、CMDI评分差异显著,存活率高,炎症适宜。固定抗原乳化液蛋白浓度及醋酸浓度,分别考察醋酸在体内滞留时间为13、15、18、20、22 s造模情况,醋酸在体内滞留时间为15 s时,模型组DAI评分、CMDI、HS评分与正常组相比差异显著,存活率高,炎症较为适宜。结论最佳UC造模条件为:5%醋酸溶液1 m L灌肠,滞留15 s后,4 m L生理盐水冲洗,再以1 m L浓度为8 mg/m L的抗原乳化液蛋白灌肠,30 min后,4 m L生理盐水冲洗。造模后的溃疡性结肠炎模型成功率高,重复性好,死亡率低,大鼠结肠炎症严重程度适宜,且病变机制与人类UC相符,此复合型溃疡性结肠炎模型适合用于UC相关研究。
Objective To optimize and evaluate the establishment of a rat model of allogeneic antigen combined with acetic acid induced ulcerative colitis. The ulcerative colitis model after modeling has high success rate, good repeatability, low mortality, suitable severity of colonic inflammation in rats, and the pathological mechanism is consistent with human UC, and this compound ulcerative colitis model is suitable for UC related the study.
