目的:探讨熊果酸对人胃癌细胞BGC823的增殖抑制和诱导凋亡作用及其机制。方法:运用MTT法检测细胞的增殖;Hoechest33258荧光染色观察DNA片段化;流式细胞术分析细胞周期和凋亡率;Westernblot检测BGC823细胞内细胞色素C(cytochromeC,cytC)、survivin、caspase3的表达。结果:熊果酸对BGC823细胞具有抑制增殖和诱导凋亡作用,并呈浓度和时间依赖性,作用24h的IC50为43.10μmol·L-1;在熊果酸作用下BGC823细胞呈凋亡征象;G1期细胞数减少,细胞主要阻滞在S期;同时cytC释放和caspase3酶原活化增加,survivin表达降低。结论:熊果酸能够在体外抑制人胃癌细胞BGC823增殖并诱导其凋亡,而促进cytC释放增加,下调survivin表达,继而引起caspase3的活化可能是其作用机制之一。
Objective: To investigate the effect of ursolic acid on the proliferation and apoptosis of human gastric cancer cell line BGC823 and its mechanism. Methods: The proliferation of BGC823 cells was detected by MTT assay. DNA fragmentation was observed by Hoechest33258 staining. Cell cycle and apoptosis rate were analyzed by flow cytometry. The expressions of cytochrome C, cytochrome C, survivin and caspase3 in BGC823 cells were detected by Western blot. Results: Ursolic acid inhibited proliferation and induced apoptosis of BGC823 cells in a dose- and time-dependent manner. The IC50 value of BGC823 cells was 43.10 μmol·L-1 for 24 h. The apoptosis of BGC823 cells was observed under the action of ursolic acid. G1 phase cells decreased, the cells mainly in the S phase; at the same time cytC release and activation of caspase 3 increased, survivin expression decreased. Conclusion: UA can inhibit the proliferation and induce the apoptosis of BGC823 cells in vitro, and promote the release of cytC, down-regulate the expression of survivin and then induce the activation of caspase3, which may be one of its mechanisms.
