目的:探讨配对同源结构域基因1(PITX1)和人端粒酶逆转录酶(hTERT)基因mRNA在非小细胞肺癌(NSCLC)组织中的表达水平与临床病理因素的关系及其相关性。方法:两步法RT-PCR技术检测91例NSCLC组织和21例支气管扩张或肺大疱等手术切除的良性肺组织中PITX1和hTERT基因mRNA的表达水平。结果:NSCLC组织PITX1基因阳性表达率为24.18%(22/91),明显低于良性肺组织的66.67%(14/21),差异有统计学意义,χ2=14.12,P=0.000。91例NSCLC组织中,hTERT基因过表达72例(79.12%),明显高于其在良性肺组织中的正常表达,χ2=46.52,P=0.000。NSCLC组织PITX1基因mRNA表达与淋巴结有无转移及分化程度(高、中分化与低分化)有关,P值分别为0.004和0.030。hTERT基因mRNA的过表达在不同病理类型中差异有统计学意义,P=0.003。PITX1与hTERT基因的mRNA表达存在相关性,P=0.000。结论:PITX1基因的表达可能对hTERT基因的过表达具有重要的抑制作用;PITX1和hTERT基因mRNA表达状况可能与NSCLC的发生与发展有关。
Objective: To investigate the relationship between the expression of paired homologous domain gene 1 (PITX1) and human telomerase reverse transcriptase (hTERT) mRNA in non-small cell lung cancer (NSCLC) and its clinicopathological factors. Methods: Two-step RT-PCR was used to detect the mRNA expression of PITX1 and hTERT in 91 NSCLC tissues and 21 benign lung tissues such as bronchiectasis or bullae. Results: The positive rate of PITX1 gene in NSCLC tissues was 24.18% (22/91), which was significantly lower than that in benign lung tissues (66.67%, 14/21), the difference was statistically significant (χ2 = 14.12, P = 0.000.91 NSCLC Tissues, hTERT gene over-expression in 72 cases (79.12%), was significantly higher than its normal expression in benign lung tissue, χ2 = 46.52, P = 0.000. NSCLC tissue PITX1 mRNA expression and lymph node metastasis and the degree of differentiation (high, moderately differentiated and poorly differentiated), P values were 0.004 and 0.030. The overexpression of hTERT gene in different pathological types was statistically significant, P = 0.003. There was a correlation between PITX1 and mRNA expression of hTERT gene, P = 0.000. CONCLUSION: The expression of PITX1 gene may have an important inhibitory effect on the overexpression of hTERT gene. The mRNA expression of PITX1 and hTERT may be related to the occurrence and development of NSCLC.
