脾虚证大鼠骨骼肌变化及中医补脾类方的作用比较

目的观察脾虚证模型大鼠骨骼肌肌力变化及中医补脾类方的作用。方法雄性Wistar大鼠随机分为正常对照组和脾虚模型组,模型组采用过度疲劳+饮食失节的方法造模4周。造模2周后模型组大鼠随机分为脾虚模型组、中药干预组(健脾升阳的补中益气汤组、健脾的四君子汤组、健脾渗湿的参苓白术散组);于造模第15天,中药各组大鼠分别按4.05 g/(kg·d)给予各干预方(补中益气汤、四君子汤、参苓白术散)灌胃;模型组和对照组给予等量蒸馏水,连续14 d。测定各组大鼠的抓握力,血液和骨骼肌相关生化指标,观察骨骼肌病理变化。结果与正常组相比,脾虚模型组大鼠抓握力、骨骼肌琥珀酸脱氢酶(SDH)、肌酸激酶同工酶(CK-MM)、肌肉素(Musclin)、胆碱酯酶受体(M-AChRM2)及Ca2+-ATPase均显著性降低,血胆碱酯酶(Ach E)显著性升高(P<0.01或P<0.05);骨骼肌出现炎细胞浸润,肌纤维排列紊乱,粗细不匀,肌丝松散及肌纤维空泡样变。与模型组相比,3个干预方组大鼠的上述指标均见不同程度地改善(P<0.01或P<0.05),骨骼肌病变减轻;其中补中益气汤组的抓握力和骨骼肌SDH、CK-MM、M-AChRM2较四君子汤组和参苓白术散组显著性升高(P<0.01或P<0.05)。结论脾虚证模型大鼠存在骨骼肌的损伤,健脾、健脾渗湿、健脾升阳3方对该模型大鼠骨骼肌均有不同程度的改善作用,但以健脾升阳的补中益气汤作用最佳。 Objective To observe the changes of skeletal muscle strength in rats with spleen deficiency syndrome and the effect of Chinese herbs for invigorating the spleen. Methods Male Wistar rats were randomly divided into normal control group and spleen deficiency model group. The model group was induced by fatigue and eating disorders for 4 weeks. Two weeks after modeling, the rats in the model group were randomly divided into spleen deficiency model group, Chinese medicine intervention group (Buzhongyiqi Decoction for invigorating the spleen and invigorating the spleen, Sijunzi Decoction for invigorating the spleen, and Shenling Baizhu Powder for invigorating the spleen and invigorating the spleen) ; On the 15th day of modeling, the rats in each group were given intragastric administration of Buzhong Yiqi Decoction, Sijunzi Decoction and Shenling Baizhu Powder at the rate of 4.05 g / (kg · d), respectively. Results Compared with the normal group, the grasping ability, the content of succinate dehydrogenase (SDH), creatine kinase (CK-MM), musclin, cholinesterase (M-AChRM2) and Ca2 + -ATPase were significantly decreased, AchE was significantly increased (P <0.01 or P <0.05); inflammatory cell infiltration was found in skeletal muscle, Uniform, loose silk and muscle fiber vacuolar degeneration. Compared with the model group, the above three indexes in the three intervention groups improved to some extent (P <0.01 or P <0.05), and alleviated the skeletal muscle lesion. Among them, the grasping power of the Zhongzhong Yiqi Decoction group and the skeletal muscle SDH, CK-MM and M-AChRM2 were significantly higher than those of Sijunzi Decoction and Shenling Baizhu Powder (P <0.01 or P <0.05). Conclusion Spleen deficiency syndrome exists skeletal muscle injury in rats. Spleen, spleen and phlegm-dampness, spleen-Yang-Yang 3 can improve the skeletal muscle of this model rat to varying degrees. However, Yiqi Tang best.
