本实验用琥珀酸脱氢酶(SDH)染色结合微机图像分析技术对尾部悬吊模拟失重90d 大鼠股直肌纤维的类型,横截面积和组织化学反应进行了定量分析。结果表明,悬吊组股直肌Ⅰ型和ⅡA 型纤维的横截面积较对照组明显缩小(P<0.01)。悬吊组股直肌Ⅰ型纤维比例显著下降(P<0.01),ⅡA 型纤维比例升高(P<0.05)。组化反应定量结果则显示:悬吊组股直肌Ⅰ型纤维SDH 含量较对照组显著减少(P<0.05),而肌糖原、肌脂类和碱性磷酸酶含量增加(P 值分别<0.01,<0.01和<0.05);悬吊组股直肌ⅡB 型纤维脂类含量增加(P<0.05),肌糖原、SDH 和ALP 无明显变化(P>0.05)。本工作的主要发现是,为期90d 的长期模拟失重可使大鼠股直肌纤维比例发生变化,氧化功能下降,并出现糖原和脂类物质蓄积。
In this study, succinic dehydrogenase (SDH) staining combined with computer image analysis of tail suspension simulated weightlessness 90d rat strangular muscle fiber type, cross-sectional area and histochemical reactions were quantitatively analyzed. The results showed that the cross-sectional area of type Ⅰ and type ⅡA fibers in the rectus femoris was significantly reduced compared with the control group (P <0.01). The proportion of type Ⅰ fibers in rectus muscle of suspension group decreased significantly (P <0.01), and the proportion of type ⅡA fibers increased (P <0.05). The quantitative results of the histochemical reaction showed that SDH content in rectus femoris typeⅠfibers in suspension group was significantly lower than that in control group (P <0.05), while muscle glycogen, myo-lipids and alkaline phosphatase contents were increased (P < 0.01, <0.01 and <0.05). The content of type IIB fibrinolipid increased in the rectus femoris group (P <0.05), but there was no significant change in muscle glycogen, SDH and ALP (P> 0.05). The main finding of this work is that the long-term simulated weightlessness of 90 days can change the ratio of the strangular rectus femoris fibers in rats, and the oxidative function decreases, and the accumulation of glycogen and lipids appear.
