目的:调查广西南宁地区人群是否存在β纤维蛋白原基因-455G/A多态性及其与纤维蛋白原表达的关系,并与其他地区比较。方法:调查于2003-11/2004-05在广西医科大学第一附属医院进行,应用聚合酶联反应-限制性片断长度多态性(PCR-RFLP)技术检测南宁地区123份DNA的β纤维蛋白原基因-455G/A多态性,用PT衍生法测定全部123份血浆纤维蛋白原水平;检索国内及欧洲部分文献中β纤维蛋白原基因-455G/A基因型频率后与之比较。结果:检出β纤维蛋白原基因-455G/A多态性,GG型为常见基因型,G/A等位基因频率为0.789/0.211。基因频率与欧洲部分国家相近(P>0.05),但在国内存在差异。A等位基因组纤维蛋白原量为(3.4585±0.7231)g/L,G等位基因组纤维蛋白原量为(3.5058±0.7171)g/L,两等位基因组纤维蛋白原量比较差异无显著性意义(P=0.7251)。结论:南宁地区多人群存在β纤维蛋白原基因-455G/A多态性,该基因多态性可能存在一定的地域性差异。G/A等位基因对血浆纤维蛋白原水平的影响不大。
Objective: To investigate whether there is a -455G / A polymorphism of β-fibrinogen gene and its relationship with fibrinogen in Nanning population of Guangxi and compare with other regions. Methods: The survey was performed at the First Affiliated Hospital of Guangxi Medical University from November 2003 to May 2004 with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) to detect 123 DNA β-fiber in Nanning The original gene -455G / A polymorphism was determined by PT method. All 123 plasma fibrinogen levels were compared with those of the domestic and European literature, and the frequencies of -455G / A genotype of β-fibrinogen gene were searched. Results: The -455G / A polymorphism of β-fibrinogen gene was detected. The GG genotype was common, and the G / A allele frequency was 0.789 / 0.211. The gene frequency was similar to that in some European countries (P> 0.05), but there were differences in China. (3.4585 ± 0.7231) g / L for the A allele and (3.5058 ± 0.7171) g / L for the G allele, there was no significant difference between the two alleles in the amount of fibrinogen (P = 0.7251). CONCLUSION: There is polymorphism of β-fibrinogen gene -455G / A in Nanning population. There may be some regional differences in the polymorphism of this gene. G / A allele has little effect on plasma fibrinogen level.
