本文在人二倍体细胞培养上就影响干扰素超诱导的某些因素进行了研究。结果表明:人肌皮二倍体细胞株的干扰素超诱导能力高于人肺二倍体细胞株。黄芪处理细胞在超诱导系统中可以生产较高量的干扰素。聚I:C和环已亚胺能在超诱导的整个过程持续存在,但放线菌素D和光辉霉素则在超诱导终止前作用1小时最适宜。光辉霉素与放线菌素D有相似的干扰素超诱导作用。聚I:C与环已亚胺的适宜作用时间为7~9小时。在适宜条件下,干扰素的产量在超诱导系统中比单诱导增加170~248倍。本研究可为大量生产纤维细胞干扰素及提取干扰素mRNA提供依据,并为阐明干扰素超诱导机理提供基础。
In this paper, some factors affecting the induction of interferon are studied in human diploid cell culture. The results showed that the human intermuscular diploid cell strain had higher interferon-induced ability than the human lung diploid cell line. Astragalus treated cells can produce higher amounts of interferon in the superinduction system. Poly I:C and ring imine can continue to exist in the whole process of super induction, but actinomycin D and diammycin are most suitable for 1 hour before termination of super induction. Mithramycin and actinomycin D have similar interferon super-induction. The suitable working time of poly I:C and cycloimine is 7-9 hours. Under appropriate conditions, the yield of interferon was increased 170-248 times more than that of the single induction in the superinduction system. This study can provide basis for the large-scale production of fibroblast interferon and the extraction of interferon mRNA, and provide the basis for elucidating the mechanism of interferon superinduction.
