目的:检测新生隐球菌中的热休克蛋白70(Hsp70)在不同的温度条件下表达量的差别,以探讨Hsp70在新生隐球菌致病中的作用。方法:采用鼠抗人、猪、狗等的Hsp70抗体通过Western印迹法检测6株临床分离株及1株CZYD2标准株新生隐球菌的 Hsp70,根据实验信号强弱定性判断在不同刺激条件下的 Hsp70产量。结果:在42℃×30min热应激及37℃×48h持续体外培养下新生隐球菌Hsp70产量较30℃培养下为高。结论:结合较难治愈病例分离株的Hsp70表达量高、热休克反应强的情况,推测Hsp70可能是新生隐球菌致病的一个因素。
Objective: To detect the difference of expression of heat shock protein 70 (Hsp70) in Cryptococcus neoformans under different temperature conditions in order to investigate the role of Hsp70 in the pathogenesis of Cryptococcus neoformans. Methods: The Hsp70 antibody of mouse anti-human, pig and dog was used to detect the Hsp70 of 6 clinical isolates and 1 CZYD2 standard strain Cryptococcus neoformans by Western blot. According to the signal intensity of the experiment, Hsp70 Yield. Results: The yield of Cryptococcus neoformans Hsp70 was higher than that at 30 ℃ under heat stress of 42 ℃ for 30min and continuous in vitro at 37 ℃ for 48h. Conclusion: In combination with the high expression of Hsp70 and strong heat shock response in the harder cases, it is speculated that Hsp70 may be a causative agent of Cryptococcus neoformans.
