目的观察消渴颗粒剂对高糖刺激后系膜细胞增殖和转化生长因子-β1(TGF-β1)表达的影响。方法培养正常大鼠系膜细胞,高浓度葡萄糖刺激系膜细胞,加入中药含药血清后,用MTT法和激光共聚焦显微镜分别观察系膜细胞增殖和TGF-β1的表达。结果消渴颗粒剂含药血清在48h可抑制高糖对大鼠系膜细胞的促增殖作用,在72、96 h可对抗高糖对系膜细胞增殖的抑制作用,96 h最为明显(P<0.05)。高糖刺激后TGF-β1在肾小球系膜细胞内表达增多,消渴颗粒剂明显减少系膜细胞内TGF-β1表达(P<0.05)。结论消渴颗粒剂可降低高糖致系膜细胞增殖,抑制高糖引起的TGF-β1过度表达,这可能是其防治糖尿病肾病的作用机制。
Objective To observe the effect of Xiaoke granules on the proliferation of mesangial cells and the expression of transforming growth factor-β1 (TGF-β1) after high glucose stimulation. Methods Mesangial cells were cultured in normal rats. Mesangial cells were stimulated with high concentration of glucose. After adding serum containing traditional Chinese medicine, the proliferation of mesangial cells and the expression of TGF-β1 were observed by MTT assay and confocal laser scanning microscopy. Results Xiaoyao granule drug-containing serum inhibited the effect of high glucose on rat mesangial cell proliferation at 48 h, and inhibited the effect of high glucose on the proliferation of mesangial cells at 72 h and 96 h, with the most obvious effect at 96 h (P< 0.05). After high glucose stimulation, the expression of TGF-β1 increased in mesangial cells, and Xiaoke granules significantly reduced the expression of TGF-β1 in mesangial cells (P<0.05). Conclusion Xiaoke granule can reduce the proliferation of mesangial cells induced by high glucose and inhibit the overexpression of TGF-β1 induced by high glucose, which may be the mechanism of action of preventing and treating diabetic nephropathy.
