目的测定弹性蛋白在正常腰椎间盘不同部位的含量,研究其与腰椎间盘功能的关系,并探讨组织内弹性蛋白的定量方法。方法分别取10个正常人腰椎间盘纤维环髓核浅深层组织,用Ⅹ型蛋白酶消化,应用高效液相色谱(HPLC)测定其水解产物中具有特征性的五肽缬-甘-缬-脯-甘(VGVPG)的含量,从而计算出腰椎间盘各部位弹性蛋白的含量。结果弹性蛋白仅占正常腰椎间盘组织干重的1.745%,纤维环浅深层和髓核浅深层四个不同部位弹性蛋白含量占该处组织干重的3.104%,2.251%,1.289%,1.113%。结论腰椎间盘是弹性蛋白含量很少的组织,弹性蛋白在腰椎间盘内分布极不均匀,此分布特点与其在椎间盘内的整体功能相适应。应用Ⅹ型蛋白酶消化法结合高效液相色谱技术不失为精确定量弹性蛋白的有效方法。
Objective To determine the content of elastin in different parts of normal lumbar intervertebral disc and study its relationship with lumbar intervertebral disc function and to explore the quantitative method of elastin in tissue. Methods The normal tissues of nucleus pulposus of lumbar intervertebral disc from 10 normal individuals were digested with Ⅹ - type protease. The content of pentapeptide - valine - valine - propionate in its hydrolyzate was determined by high performance liquid chromatography (HPLC) Gan (VGVPG) content, thereby calculating the various parts of the lumbar disc elastin content. Results Elastin accounted for only 1.745% of the normal dry weight of lumbar intervertebral disc tissue. The content of elastin in the superficial layers of the annulus and the superficial nucleus pulposus accounted for 3.104% and 2.251% of the dry weight of the tissue, 1.289%, 1.113%. Conclusions The lumbar intervertebral disc is a tissue with little elastic protein. The distribution of elastin in the lumbar intervertebral disc is very uneven. Its distribution is in accordance with its overall function in the intervertebral disc. Application Ⅹ type protease digestion combined with high performance liquid chromatography can be an effective method for accurate quantification of elastin.
