目的研究戈米辛J对铜离子和内皮细胞诱导的低密度脂蛋白氧化修饰的作用。方法以巴比妥酸反应物质、荧光物质及电泳迁移率测定低密度脂蛋白的氧化程度。结果戈米辛J抑制铜离子诱导的LDL氧化修饰,有效质量浓度为5μmol/L,IC50(95%可信限)为26.2(16.5~41.5)μmol/L,尚可抑制氧化的低密度脂蛋白产生的荧光物质及琼脂糖凝胶电泳迁移率;对内皮细胞诱导的低密度脂蛋白氧化修饰,戈米辛J的抑制作用呈剂量依赖关系。结论戈米辛J对铜离子和内皮细胞诱导的低密度脂蛋白氧化修饰有显著抑制作用。
Objective To study the effect of Gomisin J on the oxidative modification of low density lipoprotein induced by copper ions and endothelial cells. Methods Barbituric acid reactive substances, fluorescent substances and electrophoretic mobility were measured the degree of oxidation of LDL. Results Gomisin J inhibited copper ion-induced oxidative modification of LDL with an effective mass concentration of 5 μmol / L and IC50 (95% confidence limit) of 26.2 (16.5-41.5) μmol / L, Oxidized low-density lipoprotein-generated fluorescent material and agarose gel electrophoresis mobility; inhibition of endothelial cell-induced oxidative modification of low-density lipoprotein, Gomisin J in a dose-dependent manner. Conclusion Gomisin J has a significant inhibitory effect on oxidative modification of LDL induced by copper ions and endothelial cells.
