目的:探究细胞因子信号转导抑制因子3(SOCS-3)在鼠巨细胞病毒(MCMV)感染新生小鼠肝组织中的作用。方法:将48只小鼠随机分为对照组与病毒组(MCMV组),予MCMV组腹腔注射MCMV 20μl,3、7和14天时取标本(各8只),观察其生长发育及肝脏病理变化;ELISA法检测血清丙氨酸转氨酶(ALT),RT-PCR法和免疫组化法测肝组织的SOCS-3,并对ALT与SOCS-3行相关分析。结果:MCMV组小鼠生长发育迟缓,肝脏病变明显;ALT表达量和SOCS-3表达量明显高于对照组,差异有统计学意义(P<0.05),SOCS-3与ALT在3和7天呈正相关(P<0.05)。结论:MCMV感染可诱导SOCS-3表达增多,且在3和7天与肝脏损伤程度呈正相关。
Objective: To investigate the role of cytokine signaling inhibitor 3 (SOCS-3) in the liver of neonatal mice infected with murine cytomegalovirus (MCMV). Methods: 48 mice were randomly divided into control group and virus group (MCMV group). MCMV mice were intraperitoneally injected with 20μl MCMV, and samples were taken at 3, 7 and 14 days (8 mice each). The growth and development and pathological changes of liver were observed Serum alanine aminotransferase (ALT) was detected by ELISA, SOCS-3 in liver tissue was detected by RT-PCR and immunohistochemistry, and the correlation between ALT and SOCS-3 was analyzed. Results: The growth and development of MCMV mice were slow and the liver lesions were obvious. The expression of ALT and the expression of SOCS-3 in the MCMV group were significantly higher than those in the control group (P <0.05). SOCS-3 and ALT were detected at 3 and 7 days (P <0.05). CONCLUSIONS: MCMV infection induces an increase in SOCS-3 expression and is positively correlated with liver injury at 3 and 7 days.
