目的:观察双特异性单克隆抗体介导的人单核—巨噬细胞在裸鼠体内对肝癌生长的抑制作用。方法:用化学交联法制备双特异性单克隆抗体HAb18-MAb7及HAb18F(ab’)_2-MAb7F(ab’)_2,并将其与单核—巨噬细胞一同注入荷人肝癌裸体内,观察肿瘤体积的变化,结果:HAb18—MAb7及HAb18F(ab’)_2-MAb7F(ab’)_2治疗组裸鼠移植瘤体积的增长速度较对照组明显减慢(P<0.01),尤以后者为甚(P<0.01).结论:HAb18-MAb7及HAb18F(ab’)_2-MAb7F(ab’)_2都有明确的介导人单核—巨噬细胞抑制荷人肝癌裸鼠移植瘤生长的作用,而且后者的作用较前者强。
Objective: To observe the inhibitory effect of bispecific monoclonal antibody on the growth of human hepatoma cells in nude mice induced by human monocytes and macrophages. METHODS: Bispecific monoclonal antibodies HAb18-MAb7 and HAb18F(ab’)_2-MAb7F(ab’)_2 were prepared by chemical cross-linking and injected into nude human hepatoma cells together with monocytes and macrophages. Observing the changes in tumor volume, the results: The growth rate of transplanted tumors in nude mice with HAb18-MAb7 and HAb18F(ab’)_2-MAb7F(ab’)_2 treatment groups was significantly slower than that of the control group (P<0.01). The latter was even worse (P<0.01). CONCLUSION: Both HAb18-MAb7 and HAb18F(ab’)_2-MAb7F(ab’)_2 have clear roles in mediating the growth of human hepatocellular carcinoma in nude mice transplanted with human mononuclear-macrophages. The former is strong.
