目的检测脑源性神经营养因子BDNF对鼻咽癌细胞CNE-2迁移过程的影响,以探讨BDNF对无TrkB表达的鼻咽癌细胞促进迁移的作用及其机制。方法应用免疫印迹方法检测CNE-2细胞BDNF和TrkB蛋白的表达,采用细胞划痕实验检测BDNF对CNE-2细胞迁移能力的影响。结果 CNE-2细胞中高表达BDNF但不表达TrkB。BDNF依然能够促进CNE-2细胞迁移而且具有剂量依赖性,其中25 ng/mL BDNF促细胞迁移的作用具有明显的统计学差异(P<0.01)。K252a抑制Trk激酶活性不影响BDNF促细胞迁移作用(P<0.01)。结论 BDNF在不表达其高亲和力受体TrkB的鼻咽癌细胞中依然能够发挥促细胞迁移的作用,而且这一作用的机制很可能是通过其他受体及通路进行介导的。
Objective To investigate the effect of brain-derived neurotrophic factor BDNF on the migration of nasopharyngeal carcinoma cell line CNE-2 in order to explore the role of BDNF in promoting the migration of nasopharyngeal carcinoma cells without TrkB expression and its mechanism. Methods Western blotting was used to detect the expression of BDNF and TrkB protein in CNE-2 cells. The cell scratch assay was used to detect the effect of BDNF on the migration of CNE-2 cells. RESULTS: BDNF was highly expressed in CNE-2 cells but not in TrkB. BDNF still can promote the migration of CNE-2 cells in a dose-dependent manner. The effect of BDNF on cell migration was obvious (P <0.01). Inhibition of Trk kinase activity by K252a did not affect BDNF pro-cell migration (P <0.01). Conclusion BDNF can still promote cell migration in nasopharyngeal carcinoma cells that do not express its high-affinity receptor TrkB, and the mechanism of this effect may be mediated through other receptors and pathways.
