目的评价中药益气养阴生精方对微波辐射后雄性大鼠生殖系统相关参数的影响及睾丸组织病理形态学的变化情况。方法二级健康成年雄性Wistar大鼠54只,随机分为对照组14只,辐射组20只和中药组20只,辐射组和中药组用高功率脉冲微波辐射雄性大鼠,1周后中药组给予益气养阴生精方灌胃给药,对照组不予辐射,常规饲养。14 d后大鼠股动脉取血,分离血清,采用化学发光法检测血清性激素,并剖开附睾和附睾管,采用WL-9000精子分析仪分析精子总数和活跃精子密度,取睾丸组织光镜观察组织学变化情况。结果辐射组大鼠血清睾酮(T)浓度低于对照组,孕酮(P)浓度高于对照组,差异具有统计学意义(P<0.05);中药组的T浓度高于辐射组,P浓度低于辐射组,差异有统计学意义(P<0.05)。大鼠精子总数和活跃精子密度辐射组较对照组降低,差异有统计学意义(P<0.05);中药组的精子总数比辐射组和对照组显著提高,差异有统计学意义(P<0.05)。光镜观察大鼠睾丸组织辐射组可见生精上皮层轻度疏松,生精细胞变性、坏死和脱落,腔内精子减少,生精小管腔内间质蛋白水肿液积聚等;而经过中药灌胃治疗后大鼠睾丸生精小管腔中的精子明显密集,精子数量增高。结论高功率辐射可引起睾丸早期损伤,睾酮辐射后的变化提示睾丸间质细胞对高功率辐射较敏感;益气养阴生精方能够改善辐射后大鼠血清性激素水平,治疗后睾酮和孕激素有显著改变,可提高辐射后精子质量,改变辐射后睾丸的病理变化,从而提高其生殖能力。
Objective To evaluate the effects of Yiqi Yangyin Shengjing Prescription on reproductive system related parameters of male rats after microwave radiation and the pathological changes of testis. Methods Twenty-four healthy male adult Wistar rats were randomly divided into control group (n = 14), radiation group (n = 20) and traditional Chinese medicine group (n = 20). Radiation group and Chinese herb group Give yi yin yin spermatogenic prescription gavage administration, the control group is not radiation, conventional feeding. After 14 days, blood was collected from the femoral artery of rats and the serum was separated. The serum sex hormones were detected by chemiluminescence assay. The epididymal and epididymal tubes were dissected. WL-9000 sperm analyzer was used to analyze the total number of sperm and active sperm density. Histological changes. Results The concentration of serum testosterone (T) in the radiation group was lower than that in the control group, and the concentration of progesterone (P) in the radiation group was higher than that in the control group (P <0.05) Lower than radiation group, the difference was statistically significant (P <0.05). The total number of spermatozoa and active sperm density in irradiation group was lower than that in control group (P <0.05). The total number of sperm in TCM group was significantly higher than that in radiation group and control group (P <0.05) . Light microscope observation of rat testis tissue radiation group showed mild oligodendrocyte, spermatogenic cell degeneration, necrosis and loss, intraluminal sperm decreased, seminiferous tubule interstitial protein edema fluid accumulation; and after the traditional Chinese medicine irrigation After soothing the stomach, the spermatozoa in the seminiferous tubules of the testes of the rats were obviously dense and the number of sperm increased. Conclusion High-power radiation can cause early testicular injury. The changes of testosterone after irradiation suggest that testicular stromal cells are more sensitive to high-power radiation. Yiqi Yangyin Shengjing can improve serum levels of sex hormone in irradiated rats. After treatment, testosterone and progesterone Significant changes can improve post-radiation sperm quality, change the pathological changes of testis after radiation, thereby enhancing their reproductive capacity.
