目的探讨胎盘组织中内质网应激相关蛋白-钙联蛋白Calnexin(CNX) mRNA及蛋白表达与子痫前期发病的关系。方法剖宫产孕妇77例分为正常足月妊娠36例(A组),轻度子痫前期患者24例(B组)和重度子痫前期患者17例(C组)。RT-PCR技术检测胎盘组织CNX mRNA的表达,蛋白印迹法检测胎盘组织中CNX蛋白表达。结果 B、C组CNX mRNA表达水平高于A组[(1.3291±0.05687)(、3.4232±0.1078)vs.(0.5382±0.0345)](P<0.01);B、C组CNX蛋白表达水平高于A组[(1.5678±0.2315)、(2.1123±0.5310)vs.(0.5678±0.1089)](P<0.01)。结论 CNX参与了子痫前期的病理生理过程;内质网应激可能是子痫前期发病机制之一。
Objective To investigate the relationship between the expression of endoplasmic reticulum stress-related protein Calnexin (CNX) and the protein expression in placenta and the pathogenesis of preeclampsia. Methods Seventy-seven pregnant women with cesarean section were divided into 36 cases of normal term pregnancy (group A), 24 cases of mild preeclampsia (group B) and 17 cases of severe preeclampsia (group C). The expression of CNX mRNA in placenta was detected by RT-PCR and CNX protein in placenta was detected by Western blot. Results The expression of CNX mRNA in group B and C was higher than that in group A [(1.3291 ± 0.05687) vs (3.4232 ± 0.1078) vs. (0.5382 ± 0.0345)] (P <0.01) Group [(1.5678 ± 0.2315), (2.1123 ± 0.5310) vs. (0.5678 ± 0.1089)] (P <0.01). Conclusion CNX is involved in the pathophysiological process of preeclampsia. Endoplasmic reticulum stress may be one of the pathogenesis of preeclampsia.
