目的探讨人外周血单核细胞载脂蛋白E(Apo E)基因表达状况及其与高血压、血脂的相关性。方法分离外周血单核细胞,采用RT-PCR方法,分析120例标本外周血单核细胞Apo E基因表达,并测定其血脂总胆固醇(TC)、甘油三酯(TG)高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、载脂蛋白A1(apo A1)、载脂蛋白B100(apoB100)和脂蛋白α[Lp(α)]含量。结果Apo E基因可以在人外周血单核细胞表达,高危和极高危高血压患者外周血单核细胞Apo E基因表达水平较健康人明显下调(P<0.05),随着危险度的增高,下调更明显。Apo E基因表达水平与LDL-C呈负相关(r=-0.979,P<0.05),与TC、TGa、poB100、apoA1、Lp(α)无明显相关性(P>0.05)。结论高危和极高危高血压患者外周血单核细胞Apo E基因表达水平明显下调,并与血脂代谢异常相关联,提示Apo E基因表达水平变化与高血压的发生发展相关联。
Objective To investigate the gene expression of apolipoprotein E (Apo E) in human peripheral blood mononuclear cells (PBMC) and its relationship with hypertension and blood lipid. Methods Peripheral blood mononuclear cells were isolated and the expression of Apo E gene in peripheral blood mononuclear cells of 120 samples was analyzed by RT-PCR. The levels of total cholesterol (TC), triglyceride (HDL), high density lipoprotein C, LDL-C, apo A1, apolipoprotein B100 and lipoprotein a [Lp (α)] were measured. Results The Apo E gene was expressed in human peripheral blood mononuclear cells. The Apo E gene expression in peripheral blood mononuclear cells of high-risk and very high-risk hypertensive patients was significantly lower than that of healthy people (P <0.05). With the increase of risk, the Apo E gene expression was down-regulated More obvious. The level of Apo E gene expression was negatively correlated with LDL-C (r = -0.979, P <0.05), but not with TC, TGa, poB100, apoA1 and Lp (α) Conclusions Apo E gene expression in peripheral blood mononuclear cells is significantly down-regulated in high-risk and very-high-risk hypertensive patients and is associated with dyslipidemia, suggesting that changes in Apo E gene expression may be associated with the development of hypertension.
