目的 研究前列腺癌组织中的转化生长因子- β1 (TGF- β1)和血小板内皮细胞粘附分子(CD31 )的表达,以及与前列腺特异性抗原(PSA)、TNM分期的相关性。 方法 免疫组织化学法检测36例前列腺癌切除标本中癌组织TGF -β1蛋白的表达;CD31标记血管内皮细胞,计数肿瘤组织微血管密度(MVD);结合临床资料分析TGF -β1、MVD与PSA、TNM分期、病理分级、有无骨转移的相关性。 结果 前列腺癌及癌周组织TGF -β1表达阳性率分别为88. 9% (32 /36)和16. 7% (2 /12),P<0. 01。前列腺癌MVD平均为(76. 1±16. 6)mm2,癌周组织MVD平均为(35. 7±8. 5)mm2,两者比较差异有统计学意义(P<0. 01)。13例PSA<20ng/ml者MVD为(65. 6±9. 1)mm2, TGF -β1表达阳性率69. 2% (9 /13); 23例PSA>20ng/ml者MVD为(81. 5±12. 2)mm2,TGF- β1表达阳性率100% (23 23),两组比较差异均有统计学意义(P<0. 05)。23例TNM分期Ⅰ、Ⅱ期者MVD为( 72. 9±17. 0 )mm2,TGF β1表达阳性率82. 6% (19 /23), 13例Ⅲ、Ⅳ期者MVD为( 84. 4±17. 3 )mm2,TGF- β1表达阳性率100% (13 /13),两组比较差异均有统计学意义(P<0. 05)。 结论 TGF- β1高表达、MVD增高与PSA及肿瘤分期呈正相关。
Objective To study the expression of transforming growth factor - β1 (TGF - β1) and platelet adhesion molecule (CD31) in prostate cancer and the correlation with prostate specific antigen (PSA) and TNM staging. Methods Immunohistochemical method was used to detect the expression of TGF-β1 in 36 specimens of prostate cancerous tissue. CD31-labeled vascular endothelial cells were collected for counting the microvessel density (MVD) of tumor tissue. The clinical data of TGF-β1, MVD and PSA, TNM Staging, pathological grade, with or without bone metastasis. Results The positive rates of TGF-β1 expression in prostate cancer and peri-cancerous tissues were 88.9% (32/36) and 16.7% (2/12) respectively, P <0.01. The average MVD of prostate cancer was (76.1 ± 16.6) mm2, and the average MVD of peri-cancerous tissue was (35.7 ± 8.5) mm2, with significant difference between the two groups (P <0.01). The MVD was (65.6 ± 9.1) mm2 and the positive rate of TGF-β1 was 69.2% (9/13) in 13 patients with PSA <20ng / ml. The MVD of 23 patients with PSA> 20ng / 5 ± 12. 2) mm2, the positive rate of TGF-β1 expression was 100% (23 23). There was significant difference between the two groups (P <0.05). MVD in 23 cases with stage I and stage II TNM was (72.9 ± 17.0) mm2, the positive rate of TGF β1 expression was 82.6% (19/23). MVD in 13 cases with stage Ⅲ and Ⅳ was (84.4) ± 17.3) mm2, and the positive rate of TGF-β1 expression was 100% (13/13). There was significant difference between the two groups (P <0.05). Conclusion The high expression of TGF-β1 and the increase of MVD have positive correlation with PSA and tumor staging.
