目的:观察盐酸赖氨酸和七叶皂苷钠联用对大鼠脑出血后脑水肿及脑内TNF-α,IL-6的表达的影响。方法:制备大鼠脑出血模型,将实验大鼠随机分成四组,七叶皂苷钠组、联合治疗组、模型组和正常组。七叶皂苷钠组于给予七叶皂苷钠针,联合治疗组给予供试液,模型组给予生理盐水。干湿称重法测定各组脑组织含水量,免疫组化染色观察血肿周围脑组织TNF-α,IL-6的表达。结果:七叶皂苷钠组、联合治疗组的脑组织含水量显著低于模型组(P<0.01),但七叶皂苷钠组与联合治疗组比较差异无统计学意义(P>0.05)。免疫组化染色显示模型组血肿周围脑组织内可见TNF-α、IL-6显著表达,七叶皂苷钠组和联合治疗组的表达明显减少,联合治疗组脑内TNF-α、IL-6的表达显著低于七叶皂苷钠组(P<0.01)。结论:盐酸赖氨酸联合七叶皂苷钠治疗能显著减轻脑出血后脑水肿,其作用机理可能与抑制脑内血肿周围TNF-α,IL-6的表达有关。
Objective: To observe the effects of combined administration of lysine hydrochloride and sodium aescinate on cerebral edema and expression of TNF-α and IL-6 in brain after intracerebral hemorrhage in rats. Methods: The model of intracerebral hemorrhage in rats was prepared. The rats were randomly divided into four groups, sodium aescinate group, combination therapy group, model group and normal group. The sodium aescinate group was given sodium aescinate. The combined treatment group was given test solution, and the model group was given normal saline. The water content of brain tissue in each group was measured by wet-dry weighing method. The expression of TNF-α and IL-6 in the brain tissue around the hematoma was observed by immunohistochemical staining. Results: The content of water in the brain of the sodium aescinate group and the combined treatment group was significantly lower than that of the model group (P <0.01), but there was no significant difference between the sodium aescinate group and the combination therapy group (P> 0.05). Immunohistochemical staining showed that TNF-α and IL-6 were significantly expressed in the brain tissue around the hematoma model group, and the expression of TNF-α and IL-6 in the combined treatment group was significantly decreased The expression was significantly lower than sodium aescinate group (P <0.01). CONCLUSION: Lysine hydrochloride combined with sodium aescinate can significantly reduce brain edema after intracerebral hemorrhage. The mechanism may be related to the inhibition of the expression of TNF-α and IL-6 around the intracerebral hematoma.
