目的:观察活络健骨胶囊对大鼠佐剂性关节炎的抗炎免疫作用。方法:实验于2005-12-25/2006-01-23在湖南环境生物学院医学部药理实验室完成。①将48只wistar大鼠采用随机数字法分成6组,正常对照组和模型组:均给予等量蒸馏水;阳性对照组:给予雷公藤多甙7mg/kg;活络健骨胶囊高、中、低剂量组:剂量分别为15,10,5g/kg。活络健骨胶囊为老中医祖传经验方(由制川乌、赤芍、桂枝10多味中草药组成,原料药购于衡阳药材公司,鉴定为合格品,实验药液由本实验制备)。造模前3d开始给药,造模后连续给药25d。给药途径为灌胃,给药体积为等容15mL/kg。②建立大鼠后右足佐剂性关节炎模型。③造模后第2,5,10,15,20,25d分别测各鼠右后足容积,计算各组大鼠右后足在不同时间的肿胀度。④每周观察大鼠体质量增长率及造模后26d计算脾、肾上腺指数。增长率=(给药后体质量-给药前体质量)/给药前体质量×100%;器官指数=器官湿重/大鼠体质量×100。⑤观察大鼠佐剂性关节炎渗出液中白细胞介素2,白细胞介素6及前列腺素E2含量。结果:48只大鼠全部进入分析。①活络健骨胶囊对大鼠体质量及脾、肾上腺素的影响:活络健骨胶囊高、中剂量组能抑制造模鼠体质量的减轻(P<0.01);对肾上腺指数有促进作用(P<0.05)。②活络健骨胶囊对大鼠佐剂性关节炎足跖肿胀的影响:活络健骨胶囊各剂量组对造模足跖有抑制作用,而中、高剂量组的抑制作用更显著(P<0.01)。③活络健骨胶囊对佐剂性关节炎渗出液中白细胞介素2、白细胞介素6及前列腺素E2含量的影响:活络健骨胶囊各剂量组对大鼠佐剂性关节炎中白细胞介素6及前列腺素E2的生成均有显著抑制作用(P<0.01),而对白细胞介素2的含量无明显影响。结论:活络健骨胶囊具有良好的抗炎免疫效果,其作用机制之一可能是与抑制单核吞噬细胞的激活、分化及介导有关。
Objective: To observe the anti-inflammatory and immune effects of Huoluo Jianu Capsule on adjuvant arthritis in rats. METHODS: The experiment was performed at the Laboratory of Pharmacology, School of Medicine, Hunan Environment Biology Institute from December 2005 to January 25, 2006. 1 48 Wistar rats were divided into 6 groups by random number method. The normal control group and model group were given the same amount of distilled water; the positive control group was given Tripterygium wilfordii 7 mg/kg; Huoluo Jiangu Capsule was high, medium and low. Dosage group: The doses were 15, 10, and 5 g/kg, respectively. Huoluo Jiangu Capsule is an old-time traditional Chinese medicine ancestral experience party (made of Chuanwu, Red Peony, and Guizhi 10 multi-flavored Chinese herbal medicines. Administration was started 3 days before model establishment and 25 days after model establishment. The administration route was gavage, and the volume of administration was equal volume 15 mL/kg. 2 After the establishment of rat right adjuvant arthritis model. 3 The right hind paw volume of each rat was measured on the 2nd, 5th, 10th, 15th, 20th and 25th days after modeling, and the degree of swelling of the right hind paw was calculated at different times in each group. 4 Observe the growth rate of body weight in rats every week and calculate spleen and adrenal index at 26 days after modeling. Growth rate = (body mass after administration - body mass before administration)/body mass before administration × 100%; organ index = wet body weight/rat body mass × 100. (5) The levels of interleukin-2, interleukin-6 and prostaglandin E2 in the exudate of adjuvant arthritis rats were observed. Results: All 48 rats entered the analysis. 1 Effect of Huoluo Jiangu Capsule on body weight, spleen and adrenergic in rats: Huoluo Jiangu capsule high and middle dose group can inhibit the reduction of model rats’ body mass (P<0.01); it can promote the adrenal index (P<0.01). <0.05). 2 Effect of Huoluo Jiangu capsule on paw swelling of adjuvant arthritis in rats: The Huoxue Jianu capsule has inhibitory effects on the production of model athlete’s foot, but the inhibitory effect of the high dose group is more significant (P<0.01). ). Effect of Huoluo Jiangu Capsule on IL-2, IL-6, and PGE2 in exudate of adjuvant arthritis: The effect of Huoluo Jiangu capsule on adjuvant rat arthritis The production of prostaglandin 6 and prostaglandin E2 were all significantly inhibited (P<0.01), but had no significant effect on the level of interleukin-2. Conclusion: Huoluo Jiangu capsule has good anti-inflammatory and immune effects. One of its mechanisms may be related to the inhibition of the activation, differentiation and mediation of mononuclear phagocytes.
