用甘薯青枯病菌(Pseudomonassolanacearcum)免疫的BALB/C小鼠脾细胞与鼠骨髓瘤细胞杂交融合,建立了4株分泌抗甘薯青枯病菌的特异性、有鉴别力的杂交瘤细胞株:PSB24、PSB28、PSB46、PSB56。杂交瘤细胞株染色体数范围为98~104,ELISA试验培养液抗体滴度为102~103,腹水抗体滴度为105~106。这些McAb的免疫球蛋白的亚类分别属于IgG1和IgM。这4株单抗分别与来自不同区域的156株甘薯青枯病菌进行酶联免疫吸附试验,结果表明:可将这些菌株识别出4个抗原决定簇,区分出7个血清型(Ⅰ~Ⅶ)。在7个血清型中Ⅰ型和Ⅲ型在我省数量大、分布面广,是我省主要优势流行型;而Ⅶ型数量少、分布面小,可视为我省的一个弱势型
Four BALB / c mice immunized with Pseudomonas spolanacearcum were immunized with murine myeloma cells, and four specific and discriminant hybridoma cell lines secreting the bacterial wilt of sweet potato were established: PSB24, PSB28, PSB46, PSB56. The number of chromosomes in the hybridoma cell lines ranged from 98 to 104. The antibody titers of the hybridoma cells ranged from 102 to 103, and the antibody titer of ascites was 105 to 106. The subclasses of these McAbs, immunoglobulins, belong to IgGl and IgM, respectively. The four McAbs against 156 bacterial wilt bacterial fungi from different regions were tested for enzyme-linked immunosorbent assay (ELISA). The results showed that four antigenic determinants of these strains were identified and seven serotypes (Ⅰ-Ⅶ) . In the seven serotypes, type I and type III are large in number and widely distributed in our province, which is the predominant popular type in our province. However, the number of type Ⅶ is small and the distribution area is small, so it can be regarded as a vulnerable type in our province
